Sulfation‐Tunable Peptide‐Glycosaminoglycan Hydrogels for Hematopoietic Stem and Progenitor Cell Expansion

open access: yesAdvanced Functional Materials, EarlyView.
Co‐assembly of rationally designed glycosaminoglycan‐binding peptides with sulfated glycosaminoglycans yields supramolecular hydrogels with tunable mechanics and matrix‐mediated cytokine retention. The glycosaminoglycans thereby act as cofactors, driving a coil‐to‐β‐sheet transition that triggers gelation.

Mechanisms of thrombocytopenia in platelet-type von Willebrand disease

open access: yesHaematologica, 2019
Platelet-type von Willebrand disease is an inherited platelet disorder characterized by thrombocytopenia with large platelets caused by gain-of-function variants in GP1BA leading to enhanced GPIbα-von Willebrand factor (vWF) interaction.

From Payload‐First Toward Dual‐Mechanism Antibody–Drug Conjugates

open access: yesAdvanced Materials, EarlyView.
Conjugation converts potent antibodies into underexposed carriers. This perspective defines the antibody exposure deficit and maps a mechanism‐first design space from payload‐first to antibody‐first ADC architectures, integrating DAR, linker chemistry, and Fc engineering to guide rational design of constructs that balance targeted cytotoxicity with ...

Tetraspanin CD9 participates in dysmegakaryopoiesis and stromal interactions in primary myelofibrosis

open access: yesHaematologica, 2015
Primary myelofibrosis is characterized by clonal myeloproliferation, dysmegakaryopoiesis, extramedullary hematopoiesis associated with myelofibrosis and altered stroma in the bone marrow and spleen.

Modified application of Kawamoto’s film method for super‐resolution imaging of megakaryocytes in undecalcified bone marrow

open access: yesResearch and Practice in Thrombosis and Haemostasis, 2020
Background Super‐resolution microscopy has enabled high‐resolution imaging of the actin cytoskeleton in megakaryocytes and platelets. These technologies have extended our knowledge of thrombopoiesis and platelet spreading using megakaryocytes and ...

Anisotropic Masked Mycobacterium Potentiates Amplified Antitumor Trained Immunity via Spleen Targeting and Myelopoiesis Conversion

open access: yesAdvanced Materials, EarlyView.
An engineered Trojan Mycobacterium, developed via a “peeling‐off and masking‐up” strategy, functions as a powerful splenic myelopoiesis converter. This spleen‐targeted nanomedicine induces trained immunity to reprogram immunosuppressive myeloid reservoirs into antitumoral effectors.

Gfi1b controls integrin signaling-dependent cytoskeleton dynamics and organization in megakaryocytes

open access: yesHaematologica, 2017
Mutations in GFI1B are associated with inherited bleeding disorders called GFI1B-related thrombocytopenias. We show here that mice with a megakaryocyte-specific Gfi1b deletion exhibit a macrothrombocytopenic phenotype along a megakaryocytic dysplasia ...

Mutated JAK2 signal transduction in human induced pluripotent stem cell (iPSC)-derived megakaryocytes

open access: yesPlatelets, 2022
Janus kinase 2 (JAK2) gene mutations are the main drivers for polycythemia vera (PV) and essential thrombocythemia (ET). The mechanisms of single altered gene causing two different diseases are unclear.

Gut Microbiota Regulates Systemic Inflammatory Response and Compensatory Anti‐Inflammatory Response Syndromes by Targeting PF4+ Macrophages in Acute Pancreatitis

open access: yesAdvanced Science, EarlyView.
ABSTRACT Acute pancreatitis (AP) begins with pancreatic local inflammation, leading to the onset of systemic inflammatory response syndrome (SIRS), followed by compensatory anti‐inflammatory response syndrome (CARS), which causes immune paralysis and higher mortality rate.
