Mildronate exerts acute anticonvulsant and antihypnotic effects

Behavioural Pharmacology, 2010
The effects of mildronate [3-(2,2,2-trimethylhydrazinium) propionate], an inhibitor of L-carnitine biosynthesis and an anti-ischaemic drug, were examined in various in-vivo conditions to investigate the neuropharmacological profile after acute administration.

Single- and Multiple-dose Pharmacokinetic, Safety and Tolerability Study of Mildronate Injection in Healthy Chinese Subjects Pharmacokinetic of Mildronate Injection

Drug Research, 2015
Mildronate is an agent for cardioprotection and neuroprotection. This study aimed to evaluate the pharmacokinetic (PK) profiles, safety and tolerability of mildronate injection after single escalating doses and multiple doses in healthy Chinese subjects. We performed a randomized, open-label, single- and multiple-dose phase I trial including 3 doses of

Mildronate Cardioprotective Action through Carnitine-Lowering Effect

Trends in Cardiovascular Medicine, 2002
Mildronate [3-(2,2,2-trimethylhydrazinium)propionate dihydrate] ameliorates cardiac function during ischemia by modulating myocardial energy metabolism. Biochemical and pharmacological evidence supports the hypothesis that the mechanism of action of mildronate is based on its regulatory effect on carnitine concentration, whereby mildronate treatment ...

Vasorelaxant and neuroprotective effects of mildronate in a rabbit subarachnoid hemorrhage model

Turkish Neurosurgery, 2018
To investigate the effects of an anti-ischemic agent, mildronate, on subarachnoid hemorrhage-induced vasospasm.Rabbits were randomly divided into four groups: control, subarachnoid hemorrhage (SAH), vehicle, and mildronate (n=8 animals per group). In the treatment group, 200 mg/kg of mildronate were intraperitoneally administered 5 minutes after the ...

Mildronate: Mechanisms of action and prospects for correction of pathologic states

Pharmaceutical Chemistry Journal, 1995
The energy metabolism of fatty acids (FA) is attributed to their intramitochondrial [?,-oxidation. FA with number of carbon atoms less than 12 penetrate into mitochondria by a camitine-independent pathway. Carnitine-y-trimethylamino[3-hydroxybutyrate is needed for the oxidation of long-chain FA [1, 2].

The Carnitine Transporter SLC22A5 Is Not a General Drug Transporter, but It Efficiently Translocates Mildronate

Drug Metabolism and Disposition, 2009
In addition to its function as carnitine transporter, novel organic cation transporter type 2 (OCTN2; human gene symbol SLC22A5) is widely recognized as a transporter of drugs. This notion is based on several reports of direct measurement of drug accumulation. However, a rigorous, comparative, and comprehensive analysis of transport efficiency of OCTN2

Dehydration of mildronate dihydrate: a study of structural transformations and kinetics

CrystEngComm, 2014
The dehydration of mildronate dihydrate (3-(1,1,1-trimethylhydrazin-1-ium-2-yl)propionate dihydrate) was investigated by powder X-ray diffraction, thermal analysis, hot-stage microscopy, water sorption–desorption studies and dehydration kinetic studies.

[Anti-atherosclerotic action of mildronate in experiment].

Patologicheskaia fiziologiia i eksperimental'naia terapiia, 2002
Mildronate is a new antiischemic drug which inhibits biosynthesis of carnitine from butyrobetaine. We studied antiatherosclerotic and antiinflammatory effects of mildronate in guinea-pigs, rats and rabbits. We found a hypolipidemic action of this substance in rats with triton WR-1339 hyperlipidemia. A protective antiatherosclerotic effect of mildronate

[Absence of mutagenic and carcinogenic properties in Mildronate].

Voprosy onkologii, 1999
Mutagenic and carcinogenic properties of mildronate (3-[2.2.2.-trimethyl hydrazonium]-propionate) have been studied for its marked immuno-stimulating and anti-ischemic action. When dosage up to 1,000 mg/dish was used no reversal of base-pair substitution or frame shift in S.typhimurium was observed. In drosophila females treated with mildronate, mosaic

[Mildronate in the treatment of transitory ischemic attacks].

Zhurnal nevrologii i psikhiatrii imeni S.S. Korsakova, 2013
An aim of the present study was to evaluate the treatment efficacy of the antioxidant mildronate in patients with transitory ischemic attacks. We studied the dynamics of clinical status, psychometric data and indices of free-radical lipid oxidation in 40 patients. The improvement in the subjective state, memory and attention was seen in 24 patients.
