Psoriasis‐like inflammation induces structural and functional changes in mitochondria
Psoriasis‐like inflammation, induced by IL‐17, IL‐22, and TNF‐α in keratinocytes and fibroblasts, alters mitochondrial functions, suppressing respiration and increasing mitochondrial ROS production. It also triggers mitochondrial fragmentation, swelling, and cristae disassembly, alongside enhanced secretion of proinflammatory mediators driven by ...

A comprehensive overview of yeast libraries and their role in advancing cell biology
Systematic, genome‐wide yeast mutant libraries have revolutionized systems and cell biology. By allowing high‐throughput interrogation of every gene and protein, they have defined gene essentiality, charted protein localization, illuminated function, mapped interactions, and more.

Atovaquone is an antimalarial requiring potentiation for sufficient efficacy. We pursued strategies to enhance its activity, showing that 4‐nitrobenzoate inhibits 4‐hydroxybenzoate polyprenyltransferase, decreasing ubiquinone biosynthesis. Since atovaquone competes with ubiquinol in mitochondria, 4‐nitrobenzoate facilitates its action, potentiating ...

Cells must clear mislocalized or faulty proteins from membranes to survive. The AAA+ ATPase Msp1 performs this task, but dissecting how its six subunits work together is challenging. We engineered linked dimers with varied numbers of functional subunits to reveal how Msp1 subunits cooperate and use energy to extract proteins from the lipid bilayer ...

CCT4 promotes tunneling nanotube formation
Tunneling nanotubes (TNTs) are membranous tunnel‐like structures that transport molecules and organelles between cells. They vary in thickness, and thick nanotubes often contain microtubules in addition to actin fibers. We found that cells expressing monomeric CCT4 generate many thick TNTs with tubulin.
