ER proteostasis meets mitochondrial function: contact sites as hubs of communication and therapeutic targets

open access: yes
: Proteostasis maintains the balance between protein synthesis, folding, and degradation within the endoplasmic reticulum (ER). This quality-control system ensures that proteins undergo proper post-translational modifications-such as PDI-ERO1-mediated ...

Functional and transcriptomic analyses in Neurospora crassa reveal a crucial role for the N‐glycoprotein deglycosylation process in fungal homeostasis

open access: yesThe FEBS Journal, EarlyView.
In Neurospora crassa, a cytosolic GH18 endo‐β‐n‐acetylglucosaminidase (ENGase) functions as an active deglycosylating enzyme. Deletion of this severely alters tolerance to multiple stress conditions and causes extensive transcriptomic reprogramming, including upregulation of a GH20 exo‐β‐n‐acetylhexosaminidase under ER stress, indicating that N. crassa

Tankyrase‐2 regulates adipocyte differentiation through AMPK/mTOR signaling

open access: yesThe FEBS Journal, EarlyView.
Tankyrase 2 (TNKS2), a PARP family member, underpins adipocyte differentiation. TNKS2 controls LKB1 protein level, and consequently, the activity of the mTOR/AMPK system and adipocyte differentiation through a layered process. Pharmacological inhibition of TNKS2 leaves other PARPs and DNA repair active and therefore represents a novel target in ...

Structures of the UBR4 complex reveal a giant arena to capture diverse substrates for ubiquitin chain elongation

open access: yesThe FEBS Journal, EarlyView.
This review summarizes recent structures of the UBR4 complex, a giant E4 ubiquitin ligase which recognizes proteins with pre‐existing ubiquitin modifications and marks them for degradation. The UBR4 complex uses a large arena lined with substrate interaction modules to select features on ubiquitinated substrates, such as the presence of N‐degrons or ...

Newly identified human aminoacyl‐tRNA synthetase complex interacting multifunctional protein 2 (AIMP2) loss‐of‐function mutations cause neurodevelopmental defects linked to cell death in a zebrafish model

open access: yesThe FEBS Journal, EarlyView.
Human AIMP2 mutations lead to severe neurodevelopmental defects and brain atrophy. Using patient‐derived fibroblasts from two individuals, we show decreased AIMP2 protein levels and overall protein synthesis. In a zebrafish loss‐of‐function model, the lack of AIMP2 leads to an increase in cell death and results in smaller brains.

Selective autophagy pathways in ageing and neurodegeneration: Emerging roles for mitophagy and nucleophagy

open access: yesThe FEBS Journal, EarlyView.
As we age, cells gradually accumulate damage, which can contribute to disease and neurodegeneration. This review explores how two cellular recycling processes, mitophagy and nucleophagy, remove damaged mitochondria and nuclear material, respectively, to maintain cell health and long‐term neuronal homeostasis.

The transport of protein aggregates to the vacuole suggests a role for lipid droplets in waste disposal in Saccharomyces cerevisiae

open access: yesThe FEBS Journal, EarlyView.
Lipid droplets (LD) act as cellular ‘garbage trucks’ that transport cytosolic protein aggregates to the vacuole for degradation in yeast. Protein aggregates are docked to lipid droplets in a triacylglycerol‐dependent manner, transported through the cytoplasm, and transferred to the insoluble protein deposit (IPOD) at the vacuole.

Proteostasis in the regulation of pancreatic islet cell plasticity

open access: yesJournal of Diabetes Investigation, EarlyView.
Adaptive proteostasis integrates protein synthesis, folding, trafficking, and degradation through ISR, UPR, ER stress responses, autophagy, and proteasomal degradation. In pancreatic islet cells, this coordinated network preserves protein quality, secretory function, proliferation, and survival, enabling adaptation to metabolic stress and aging while ...
