Downregulation of MLL1 Promotes Intestinal Epithelial Barrier Repair Through Gata4/Bmp4 Activation to Ameliorate Crohn's Disease‐Like Colitis in Mice

open access: yesCell Proliferation, EarlyView.
This study elucidates the pivotal regulatory role and mechanism of MLL1 in inflammatory bowel disease. Experimental data demonstrated elevated MLL1 expression levels in TNBS‐induced murine colitis models, with pharmacological inhibition of MLL1 significantly promoting recovery from CD‐like colitis by accelerating intestinal barrier restoration ...

Concentration dependent chromatin states induced by the bicoid morphogen gradient

open access: yeseLife, 2017
In Drosophila, graded expression of the maternal transcription factor Bicoid (Bcd) provides positional information to activate target genes at different positions along the anterior-posterior axis.

Scaling of morphogen gradients

open access: yesCurrent Opinion in Genetics & Development, 2011
Individuals of the same or closely related species can vary substantially in size. Still, the proportions within and between tissues are precisely kept. This adaptation of pattern with size termed scaling, is receiving a growing attention. We review experimental evidence for scaling, and describe theoretical models for mechanisms that scale morphogen ...

Irradiated Liver Cancer Cell Vaccine Transfected With GM‐CSF Induces Specific and Long‐Lasting Anti‐tumour Immunity Through the Synergistic Effect of Oxidised mtDNA and GM‐CSF

open access: yesCell Proliferation, EarlyView.
Song et al. developed an mGM‐CSF‐loaded, liver cancer cell‐based vaccine that potently activated dendritic cells and elicited tumour‐specific T cell responses. Mechanistically, radiation‐induced oxidative damage to mitochondrial DNA in cancer cells triggers the cGAS‐STING pathway in DCs, which synergizes with mGM‐CSF‐driven JAK2‐STAT3 signalling to ...

Extracellular matrix and proteolysis: mechanisms driving irreversible changes and shaping cell behavior

open access: yesThe FEBS Journal, EarlyView.
Irreversible ECM proteolysis by remodeling enzymes shapes development, homeostasis, and disease. ECM‐degrading proteases display cell specificity and are governed by shared mechanisms, exhibiting functional redundancy in generating matrikines, growth factors, and cytokines.

Proteolysis at the extracellular matrix interface: Molecular architects and regulators in health and disease

open access: yesThe FEBS Journal, EarlyView.
The extracellular matrix (ECM) is a dynamic scaffold that orchestrates tissue architecture and cellular communication. A critical but underexplored interplay between proteases and cluster of differentiation molecules (CD) governs ECM turnover and directs cell fate.

Rapid changes in morphogen concentration control self-organized patterning in human embryonic stem cells

open access: yeseLife, 2019
During embryonic development, diffusible signaling molecules called morphogens are thought to determine cell fates in a concentration-dependent way.

Orchestrated molecular changes of proliferative, migratory‐fibrillar, synaptic, and postmigratory compartments align with precocious cortex‐type specification in the early human pallium

open access: yesJournal of Anatomy, EarlyView.
Early human cortical development is organized by transient cellular compartments that define cortical types before mature layers form. Analysis of the human fetal pallium (7.5–15 PCW) shows distinct spatiotemporal trajectories for the archicortex, mesocortex, and neocortex, with delayed but accelerated differentiation in allocortical regions.

Morphogen Gradients, in Theory [PDF]

open access: yesDevelopmental Cell, 2002
The idea that morphogen gradients are established by a process of repeated cycles of exocytosis and endocytosis-so-called planar transcytosis-has been gaining acceptance. This is now challenged by a theoretical approach that experimental biologists should not dismiss; diffusive mechanisms of gradient formation may be correct after all.

Built different: ER cisternae formed by the Arabidopsis Lunapark proteins differ in ultrastructure and affect ER–Golgi transport

open access: yesNew Phytologist, EarlyView.
The endoplasmic reticulum (ER) is a key structure in plant cells that helps produce and transport proteins. We discovered that different Lunapark proteins shape ER structures in distinct ways, affecting protein movement and secretion. Understanding how ER structure influences function can help improve plant‐based production of valuable compounds, such ...
