Advanced Molecular Analysis in Hemophilia A in a Single Step: Next Generation Sequencing (NGS) and Copy Number Variation (CNV) Analysis

open access: yesInternational Journal of Laboratory Hematology, EarlyView.
ABSTRACT Background Hemophilia A, an X‐linked bleeding disorder caused by pathogenic variants in the F8 gene, requires precise genetic diagnosis for optimal management. Conventional stepwise sequence and copy number variation (CNV) analyses are time‐consuming and may leave some cases unresolved.

‘It's not much, but it's honest work’: The status of environmental DNA analyses of fish biodiversity in southern Africa

open access: yesJournal of Fish Biology, EarlyView.
Abstract Environmental DNA (eDNA) biodiversity surveys have the power to transform the detection of species in natural environments, which is crucial for the conservation and management of freshwater, estuarine and marine environments. Globally, eDNA‐based analyses have increased significantly, with fishes being the most widely studied aquatic ...

Prospects of multipurpose biomonitoring for fisheries assessment based on environmental nucleic acids

open access: yesJournal of Fish Biology, EarlyView.
Abstract Methods using environmental nucleic acids have become highly effective for monitoring aquatic biodiversity, with an array of suitable use cases, including metrics for fisheries assessment. Traditional methods for assessing fish populations often rely on invasive techniques with limited spatial and temporal coverage.

Synergistic engineering of Casδ nuclease for robust genome editing

open access: yesJournal of Integrative Plant Biology, EarlyView.
A hierarchical engineering strategy was used to enhance the editing activity of Casδ, yielding an optimized variant termed enCasδ. This variant enables robust genome editing in animal cells and plants, with overall editing performance comparable to that of SpCas9 and other Cas12 nucleases.

20th Anniversary of human‐induced pluripotent stem cells and the role of microscopy

open access: yesJournal of Microscopy, EarlyView.
Abstract It has been 20 years since the pioneering work of Shinya Yamanaka and Kazutoshi Takahashi at Kyoto University led to the first successful generation of induced pluripotent stem cells (iPSCs) from mouse embryonic and adult fibroblast cells. iPSCs have the capacity to differentiate into any type of cell in the human body, and as such, they have ...

Transcriptomic profiling of subpopulations of mouse embryonic subplate neurons

open access: yesJournal of Anatomy, EarlyView.
To study gene expression in subpopulations of cortical subplate neurons (Lpar1‐EGFP‐positive and D1B‐positive SpN clusters), we applied gene expression profiling, including bulk microarray analysis, single‐cell RNA sequencing (scRNA‐seq), and Visium spatial transcriptomics, and identified both overlapping and unique gene expression signatures ...
