Downregulation of NAD Kinase Expression in β-Cells Contributes to the Aging-Associated Decline in Glucose-Stimulated Insulin Secretion. [PDF]

open access: yesAging Cell
Adequate NADK/NADK2 level is essential to the proper functioning of redox and folate cycles and the purine biosynthesis pathway, and as well as maintenance of GSIS response. During aging, diminished NADK/NADK2 expression alters the metabolic status quo, increasing AICAR production. The latter activates AMPK and suppresses GSIS.

De Novo Biosynthesis of Antidepressant Psilocybin in Escherichia coli. [PDF]

open access: yesMicrob Biotechnol
Our work achieved de novo synthesis of antidepressant psilocybin in a single engineered Escherichia coli cell. By constructing the P450 PsiH variants with N‐terminal hydrophobic domain modifications, improving the supply of precursor tryptophan and cofactor NADPH, enhancing the electron transfer (ET) pathway and the final methylation step, and ...

Deciphering the functional landscape of phosphosites with deep neural network

open access: yesCell Reports, 2023
Summary: Current biochemical approaches have only identified the most well-characterized kinases for a tiny fraction of the phosphoproteome, and the functional assignments of phosphosites are almost negligible. Herein, we analyze the substrate preference

The Ca2+-CaM Signaling Pathway Mediates Potassium Uptake by Regulating Reactive Oxygen Species Homeostasis in Tobacco Roots Under Low-K+ Stress

open access: yesFrontiers in Plant Science, 2021
Potassium (K+) deficiency severely threatens crop growth and productivity. Calcium (Ca2+) signaling and its sensors play a central role in the response to low-K+ stress. Calmodulin (CaM) is an important Ca2+ sensor.

Distinct dual roles of p-Tyr42 RhoA GTPase in tau phosphorylation and ATP citrate lyase activation upon different Aβ concentrations

open access: yesRedox Biology, 2020
Both the accumulation of Amyloid-β (Aβ) in plaques and phosphorylation of Tau protein (p-Tau) in neurofibrillary tangles have been identified as two major symptomatic features of Alzheimer's disease (AD).

Functional identification and subcellular localization of NAD kinase in the protozoan parasite Giardia intestinalis

open access: yesRevista Colombiana de Química, 2019
Nicotinamide adenine dinucleotide phosphate (NADP) is an essential biomolecule that participates in the redox homeostasis and synthesis of signaling compounds. NAD kinase (NADK) (EC 2.7.1.23/2.7.1.86) is the only enzyme capable of synthesizing NADP. This
