S-Palmitoylation of γ-Secretase Subunits Nicastrin and APH-1 [PDF]

open access: yesJournal of Biological Chemistry, 2009
Proteolytic processing of amyloid precursor protein (APP) by beta- and gamma-secretases generates beta-amyloid (Abeta) peptides, which accumulate in the brains of individuals affected by Alzheimer disease. Detergent-resistant membrane microdomains (DRM) rich in cholesterol and sphingolipid, termed lipid rafts, have been implicated in Abeta production ...

Generation of amyloid-β is reduced by the interaction of calreticulin with amyloid precursor protein, presenilin and nicastrin.

open access: yesPLoS ONE, 2013
Dysregulation of the proteolytic processing of amyloid precursor protein by γ-secretase and the ensuing generation of amyloid-β is associated with the pathogenesis of Alzheimer's disease.

A Scoping Review of Pathogenesis, Current Treatments, and Novel Approaches for Vitiligo

open access: yesJournal of Cosmetic Dermatology, Volume 24, Issue 10, October 2025.
ABSTRACT Background Vitiligo is a chronic skin disorder characterized by loss of melanocytes, causing depigmentation and psychological distress. Its pathogenesis is complex and multifactorial, involving genetic predisposition, autoimmune, oxidative stress, and neurogenic mechanisms.

Senescence Alters Antimicrobial Peptide Expression and Induces Amyloid‐β Production in Retinal Pigment Epithelial Cells

open access: yesAging Cell, Volume 24, Issue 9, September 2025.
Healthy RPE cells produce AMPs and AMP precursors, including amyloid precursor protein (APP) and chemerin, and protect the neuroretina from blood‐borne pathogens and maintain homeostasis. When RPE cells undergo senescence, the antibacterial activity declines, and APP undergoes amylogenesis, leading to Aβ deposition.

Nontranscriptional Role of Hif-1α in Activation of γ-Secretase and Notch Signaling in Breast Cancer

open access: yesCell Reports, 2014
γ-Secretase is composed of four proteins that are obligatory for protease activity: presenilin, nicastrin, Aph1, and Pen-2. Despite the progress toward understanding the function of these individual subunits, there is no information available pertaining ...

Cannabidiol and Alzheimer Disease: A Comprehensive Review and In Silico Insights Into Molecular Interactions

open access: yesEuropean Journal of Neuroscience, Volume 62, Issue 4, August 2025.
Cannabidiol improves several Alzheimer's‐related aspects and regulates disease‐associated genes. Literature review and Over‐Representation Analysis of 64 genes confirm its effects on key mechanisms such as amyloid‐β and tau processing, oxidative stress, inflammation, and lipid metabolism.

NCSTN promotes hepatocellular carcinoma cell growth and metastasis via β-catenin activation in a Notch1/AKT dependent manner

open access: yesJournal of Experimental & Clinical Cancer Research, 2020
Background Hepatocellular carcinoma is the third top cause of cancer-related mortalities worldwide. The prognosis of HCC patients remains poor due to rapid progression and high incidence of tumor recurrence.

Vitamin D receptor is present on the neuronal plasma membrane and is co-localized with amyloid precursor protein, ADAM10 or Nicastrin. [PDF]

open access: yesPLoS ONE, 2017
Our recent study indicated that vitamin D and its receptors are important parts of the amyloid processing pathway in neurons. Yet the role of vitamin D receptor (VDR) in amyloid pathogenesis is complex and all regulations over the production of amyloid ...

Understanding the Intersection Between Hormonal Dynamics and Brain Plasticity in Alzheimer's Disease: A Narrative Review for Implementing New Therapeutic Strategies

open access: yesHealth Science Reports, Volume 8, Issue 7, July 2025.
ABSTRACT Background and Aims Alzheimer's disease (AD), a chronic neurodegenerative disorder, is portrayed by neurocognitive decline in the structure and function of the human brain. Various factors are implicated in the pathogenesis to neuroplasticity alteration in the brain of an individual afflicted with AD.

Drosophila Nicastrin Is Essential for the Intramembranous Cleavage of Notch [PDF]

open access: yesDevelopmental Cell, 2002
The catalytic subunit of gamma-secretase is thought to be Presenilin, which is required for both the cleavage of APP and in the processing of Notch. Presenilin is found in a multisubunit complex that also contains Nicastrin. Nicastrin has been implicated in APP processing, but its role in Notch signaling remains unclear.
