Inhibition of NLRP3 Inflammasome: A Prospective Target for the Treatment of Ischemic Stroke

open access: yesFrontiers in Cellular Neuroscience, 2020
Stroke is one of the major devastating diseases with no effective medical therapeutics. Because of the high rate of disability and mortality among stroke patients, new treatments are urgently required to decrease brain damage following a stroke.

Inflammasomes in neuroinflammatory and neurodegenerative diseases [PDF]

open access: yes, 2019
Neuroinflammation and neurodegeneration often result from the aberrant deposition of aggregated host proteins, including amyloid-beta, alpha-synuclein, and prions, that can activate inflammasomes.

Lactylation‐Driven YTHDC1 Alleviates MASLD by Suppressing PTPN22‐Mediated Dephosphorylation of NLRP3

open access: yesAdvanced Science, EarlyView.
In MASLD, YTHDC1 undergoes increased lactylation and ubiquitination, reducing its expression. AARS1 mediates lactylation at lysine 565, while disrupted binding to LDHA further promotes lactylation, suppressing YTHDC1. This downregulation enhances PTPN22 mRNA stability, leading to NLRP3 dephosphorylation and activation, which exacerbates inflammation ...

NLRP2 controls age-associated maternal fertility [PDF]

open access: yes, 2016
Nucleotide-binding domain and leucine-rich repeat (NLR) proteins are well-known for their key roles in the immune system. Ectopically expressed NLRP2 in immortalized cell lines assembles an inflammasome and inhibits activation of the proinflammatory ...

Fatty acid synthesis and NLRP3-inflammasome

open access: yesOncotarget, 2015
Fatty acid synthesis is an important process of the lipogenesis [1]. Fatty acid synthesis is tightly regulated by key enzymes including fatty acid synthase (FASN) [1]. FASN and lipogenesis related enzymes are upregulated in various cancer cells and associated with oncogenic progression [1].

Postoperative Stress Accelerates Atherosclerosis Through Inflammatory Remodeling of the HDL Proteome and Impaired Reverse Cholesterol Transport

open access: yesAdvanced Science, EarlyView.
The study shows that noncardiac surgical inflammation rapidly disrupts HDL function and cholesterol efflux in mice and human patients. Impaired reverse cholesterol transport after surgery drives rapid lipid accumulation, NETosis, and apoptosis within atherosclerotic plaques.

Correcting the NLRP3 inflammasome deficiency in macrophages from autoimmune NZB mice with exon skipping antisense oligonucleotides [PDF]

open access: yes, 2016
Inflammasomes are molecular complexes activated by infection and cellular stress, leading to caspase-1 activation and subsequent interleukin-1β (IL-1β) processing and cell death.
