Chronic septic arthritis involves intracellular bacterial persistence and lipid‐immune crosstalk via the PGRN‐BMP lysosomal axis. A dual‐targeting nanoparticle system (NPs@PGRN) restores lysosomal bactericidal function, reduces bacterial burden, and reprograms macrophage immunity, offering a novel therapeutic strategy. ABSTRACT Chronic septic arthritis,

Paxillin mediates ATP-induced activation of P2X7 receptor and NLRP3 inflammasome
Background Extracellular adenosine triphosphate (ATP), a key danger-associated molecular pattern (DAMP) molecule, is released to the extracellular medium during inflammation by injured parenchymal cells, dying leukocytes, and activated platelets.

Fatty acid synthesis and NLRP3-inflammasome
Fatty acid synthesis is an important process of the lipogenesis [1]. Fatty acid synthesis is tightly regulated by key enzymes including fatty acid synthase (FASN) [1]. FASN and lipogenesis related enzymes are upregulated in various cancer cells and associated with oncogenic progression [1].

Elevated exogenous palmitic acid promotes the S‐palmitoylation of SRPK1 in endothelial cells, a dynamic process governed by ZDHHC24 and APT1. This post‐translational modification strengthens the interaction between SRPK1 and the E3 ubiquitin ligase MIB1, thereby facilitating the proteasomal degradation of SRPK1.

Reformulating Pro-Oxidant Microglia in Neurodegeneration [PDF]
In neurodegenerative diseases, microglia-mediated neuroinflammation and oxidative stress are central events. Recent genome-wide transcriptomic analyses of microglial cells under different disease conditions have uncovered a new subpopulation named ...

Role of NLRP3 Inflammasomes in Atherosclerosis
Inflammation with macrophage infiltration is a key feature of atherosclerosis. Although the mechanisms had been unclear, emerging evidence unveiled that NLRP3 inflammasomes, which regulate caspase-1 activation and subsequent processing of pro-IL-1β, trigger vascular wall inflammatory responses and lead to progression of atherosclerosis.

Inhibition of NLRP3 Inflammasome: A Prospective Target for the Treatment of Ischemic Stroke
Stroke is one of the major devastating diseases with no effective medical therapeutics. Because of the high rate of disability and mortality among stroke patients, new treatments are urgently required to decrease brain damage following a stroke.

Time‐restricted feeding (TRF) exerts protein‐dependent neuroprotective effects in an MPTP‐induced Parkinson's disease model. In casein‐fed mice, TRF improves gut barrier integrity and reduces neuroinflammation, possibly via modulation of Allobaculum and BCAAs.
