Therapeutic potential of natural products in cancer immunotherapy: Advances and challenges
This review systematically outlines the mechanisms underlying tumour immunotherapy resistance and elucidates the role of natural products in enhancing therapeutic efficacy as immunomodulatory adjuvants. Abstract Immunotherapy has emerged as a clinically pivotal approach in cancer treatment, but its application remains limited to a small subset of ...

Carrageenans, widely used food additives, disrupted intestinal epithelial integrity in a gut‐on‐a‐chip model. All types (κ‐, ɩ‐, λ‐) induced cytotoxicity, inflammation and tight junction (TJ) disruption, triggering TNF‐mediated immune responses. λ‐Carrageenan had the most severe effects, supporting the Epithelial Barrier Theory linking food additives ...

Role of NOD2 variants in spondylarthritis.
OBJECTIVE: To investigate the role of the gene NOD2 in susceptibility to, and clinical manifestations of, ankylosing spondylitis (AS). METHODS: A case-control study of NOD2 polymorphisms known to be associated with Crohn's disease (CD) (Pro(268)Ser, Arg ...

Expression and functional characterization of NOD2 in decidual stromal cells isolated during the first trimester of pregnancy. [PDF]
NOD2, one of the cytosolic proteins that contain a nuclear oligomerization domain (NOD), is a pattern recognition receptor (PRR) involved in innate immune responses to intracellular pathogens. Little is known, however, about the effect of NOD2 expression

Lactate Accelerates Early Angiogenesis and Bone Regeneration Through Macrophage M1 Polarisation
During the early stage of bone defect healing, lactate accumulates and contributes to increasing NOD1 expression by stabilising HIF1α that in turn triggers a calcium influx, which ultimately polarises macrophages towards the M1 phenotype and accelerates vascularisation of endothelial cells.

ADAM17 and its proteolytic targets in disease pathogenesis
ADAM17 as a multifunctional sheddase with contrasting roles across inflammatory, metabolic, cardiovascular, and neoplastic diseases. Through regulated activation by iRhom, iTAP/FRMD8, and tetraspanins, ADAM17 cleaves diverse membrane ligands and receptors, thereby promoting inflammation, fibrosis, obesity, insulin resistance, and tumor progression ...

Ubiquitin and ubiquitin‐like modifications in the endoplasmic reticulum stress response
Endoplasmic reticulum (ER) stress activates various proteostasis control processes, including the unfolded protein response, ribosome‐associated quality control, and ER‐associated degradation. Ubiquitin and ubiquitin‐like modifications dynamically regulate these processes to determine cell fate, promoting adaptation or inducing cell death.

Loss-of-function mutations in nucleotide-binding oligomerization domain 2 (NOD2) constitute the primary risk factors for Crohn’s disease. NOD2 is an intracellular sensor for muramyl dipeptide (MDP), a small molecule derived from the peptidoglycan layer ...

Association of the TLR4 and NOD2 Polymorphisms with Childhood Acute Lymphoblastic Leukemia
Objective:Immune activation plays a critical role in the immune response against cancer. Although several genetic factors are established with regard to the development of acute lymphoblastic leukemia (ALL), the role of immunogenic genes in ALL ...

Autoimmunity and Periodontitis
In a microbe‐driven inflammatory environment, peptidyl‐arginine deiminase (PAD) enzymes from neutrophils and Porphyromonas gingivalis citrullinate both microbial and self‐antigens. B cell presentation of citrullinated or self‐mimicking epitopes activates T cells that assist B cells in antibody isotype switching, affinity maturation, epitope spreading ...
