Subpopulations of blood monocytes in patients with generalized hypoxia

open access: yesБюллетень сибирской медицины, 2019
The aim of the work is to establish general regularities and features of differentiation of blood monocytes into 4 subpopulations in diseases associated with circulatory and respiratory hypoxia.Materials and methods.

A 3D Human Bone and Bone Marrow‐on‐a‐Chip Model for In Vitro Bone Remodeling and Immune Cell Maintenance

open access: yesAdvanced Science, EarlyView.
This study presents a human Bone (and Bone Marrow)‐on‐a‐Chip model based on native human bone scaffolds and autologous cells. Dynamic perfusion and sequential cell seeding replicate the physiological bone remodeling process in vitro, enabling the long‐term culture of functional, mature bone marrow immune subpopulations.

Single‐Cell Dissection of Therapy‐Induced Remodeling Uncovers a Fibroblast‐Driven Immunosuppressive Niche and Targetable Vulnerabilities in Lethal Prostate Cancer

open access: yesAdvanced Science, EarlyView.
Single‐cell longitudinal profiling reveals that androgen‐deprivation therapy induces a DPT+ fibroblast‐complement axis that suppresses macrophage inflammation and drives CD8+ T cell exhaustion in prostate cancer. Concurrently, resistant epithelial subpopulations persist and engage TSPAN1‐ and NRXN1‐mediated programs promoting CRPC and neuroendocrine ...

Single-cell sequencing reveals a senescent immune landscape in bone marrow lesions inducing articular cartilage damage in osteoarthritis

open access: yesBone Research
Bone marrow lesions (BML) are early signs of osteoarthritis (OA) and are strongly correlated with the deterioration of cartilage lesions. Single-cell RNA sequencing (scRNA-seq) analyses were performed on BM from non-BML and BML areas and articular ...

Targeting METTL3 Attenuates Thyroid Inflammatory Injury by Restoring Th17/Treg Balance through a YTHDC2‐m6A‐Dependent KDR/VEGFA Loop

open access: yesAdvanced Science, EarlyView.
METTL3‐mediated m6A modification stabilizes KDR/Kdr mRNA through YTHDC2, amplifying the VEGFA‐KDR feedback loop and disturbing Th17/Treg balance. This pathway promotes persistent inflammation and tissue damage in patients with AIT, and inhibiting the METTL3‐KDR axis effectively disrupts this circuit, alleviating thyroid tissue injury and disease ...

Changes in monocyte subsets are associated with an increased risk of AAA and are surrogate markers for AAA morphology in patients with late-stage disease

open access: yesFrontiers in Immunology
IntroductionMonocytes play a role in the pathology of abdominal aortic aneurysm (AAA) and can display immunophenotypic heterogeneity. Alterations in monocyte subsets are associated with cardiovascular risk, but their profile in AAA is poorly understood ...

Chiral Nanoparticles Suppress Inflammatory Infiltration to Promote Extracellular Matrix Remodeling for Ectopia Lentis Therapy

open access: yesAdvanced Science, EarlyView.
L‐cysteine‐configured chiral polyurethane nanoparticles suppress ocular inflammation and reduce extracellular matrix (ECM) degradation in ectopia lentis by regulating macrophages polarization and inhibiting nuclear factor kappa B signaling pathway. By promoting zonular fiber‐associated protein restoration and tissue repair, this minimally invasive ...

Radiotherapy Enhances the Oncolytic Efficacy of the Novel Oncolytic Herpesvirus VG161 and Amplifies Its Antitumor Immunity in Breast Cancer

open access: yesAdvanced Science, EarlyView.
RT promotes VG161 replication in BC and its immunostimulatory transgenes expression, which is mediated by the upregulation of GADD34 and HVEM caused by RT. The combination therapy with VG161 and RT increases the abundances of tumor‐infiltrating lymphocytes and elicits potent systemic antitumor immunity, thereby effectively inhibiting local tumors and ...

Single-cell transcriptome analysis reveals the dysregulated monocyte state associated with tuberculosis progression

open access: yesBMC Infectious Diseases
Background In tuberculosis (TB) infection, monocytes play a crucial role in regulating the balance between immune tolerance and immune response through various mechanisms. A deeper understanding of the roles of monocyte subsets in TB immune responses may
