Non-homologous end joining repair in Xenopus egg extract [PDF]
AbstractNon-homologous end joining (NHEJ) is a major DNA double-strand break (DSB) repair mechanism. We characterized here a series of plasmid-based DSB templates that were repaired in Xenopus egg extracts via the canonical, Ku-dependent NHEJ pathway. We showed that the template with compatible ends was efficiently repaired without end processing, in a

Aging Is a Key Driver for Adult Acute Myeloid Leukemia
Acute myeloid leukemia (AML) is a classical age‐related hematologic malignancy, and a key driver of AML is aging, which profoundly regulates intrinsic factors such as genomic instability, epigenetic reprogramming, and metabolic dysregulation, and alters bone marrow microenvironment.

Non-homologous end-joining factors ofSaccharomyces cerevisiae [PDF]
DNA double-strand breaks (DSB) are considered to be a severe form of DNA damage, because if left unrepaired, they can cause a cell death and, if misrepaired, they can lead to genomic instability and, ultimately, the development of cancer in multicellular organisms.

Complementarity of Long‐Reads and Optical Mapping in Parkinson's Disease for Structural Variants
ABSTRACT Objective Long‐read sequencing and optical genome mapping technologies have the ability to detect large and complex structural variants. This has led to the discovery of novel pathogenic variants in neurodegenerative movement disorders. Thus, we aimed to systematically compare the SV detection capabilities of OGM and ONT in Parkinson's disease.

Creep‐Induced Microstructural Evolution in an A2‐B2 Superalloy
A 27.3Ta‐27.3Mo‐27.3Ti‐8Cr‐10Al (at.%) refractory high‐entropy alloy with precipitation‐strengthened A2‐B2 microstructure was studied by creep tests at 1030°C, which demonstrate a transition in deformation mechanisms in the range of 100–150 MPa applied stress. This is associated with changes in dislocation–precipitate interactions. Relevant deformation

Non-homologous end joining (NHEJ) is a DNA repair pathway that directly ligates broken DNA ends without the need for a homologous template, whereas translesion synthesis (TLS) is a DNA damage tolerance mechanism in which specialized DNA polymerases ...

Double-strand break repair assay for non-homologous end joining using HCR.
(A) Schematic drawing of host cell reactivation assay for non-homologous end joining repair. Non-homologous end joining is assessed by cleaving the pCMS-end plasmid (Xho I/Apa I) to generate a non-religatable DSB between the promoter and the YFP-coding ...

DNA end resection requires constitutive sumoylation of CtIP by CBX4
The choice between non-homologous end-joining and homologous recombination to repair a DNA double-strand break depends on activation of the end resection machinery.
