Oligodeoxynucleotides Modified with 2O(Cysteinylaminobutyl)carbamoylethylribothymidine Residues for Native Chemical Ligation with Peptide at Internal Positions

open access: yes, 2022
We used native chemical ligation (NCL) to synthesize a 2′-O-{N-[N-(S-tert-butylthiocysteinyl)aminobutyl]carbamoylethyl} (CysBCE) ribothymidine-derived oligonucleotide to expand the variety of peptide conjugation sites, allowing the incorporation of ...

A Bioinspired Three‐Dimensional High‐Curvature Nano‐Interface Integrated Microfluidic Chip for Small Extracellular Vesicles Enrichment and Machine Learning‐Assisted Prostate Cancer Precision Diagnosis

open access: yesAdvanced Science, EarlyView.
A biotin‐modified artificial insertion peptide functionalized three‐dimensional high‐curvature‐TiO2 nano‐interface was engineered in a microfluidic chip to improve the isolation efficiency of small extracellular vesicles (sEVs). This chip balanced affinity, releasability, and extendibility, enabling high‐throughput recovery of sEVs for downstream ...

Design and Evaluation of a Novel Peptide–Drug Conjugate Covalently Targeting SARS-CoV-2 Papain-like Protease

open access: yes, 2022
Coronavirus disease 2019 (COVID-19) pandemic, a global health threat, was caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). The SARS-CoV-2 papain-like cysteine protease (PLpro) was recognized as a promising drug target because of ...

Microglia‐Targeted Biomimetic Tetrahedral Framework Nucleic Acid Nanovesicles for Synergistic Treatment of Sepsis‐Associated Encephalopathy

open access: yesAdvanced Science, EarlyView.
Sepsis‐associated encephalopathy (SAE) lacks effective therapies. We developed ME@FDsi, a biomimetic nanodrug using a tetrahedral framework nucleic acid to deliver disulfiram and siTNFα. It crosses the blood‐brain barrier, targets M1 microglia, inhibits pyroptosis and inflammation, and scavenges ROS.

Hepatoma‐Derived Growth Factor Coordinates STAT3 Pathway and Exosome‐Mediated Intrahepatic Crosstalk to Control Hepatic Steatosis and MASLD

open access: yesAdvanced Science, EarlyView.
Hepatic HDGF as a key mediator in coordinating hepatic steatosis and intrahepatic crosstalk in MASLD. Activation of HDGF facilitates its interaction with both STAT3 and S6K1, driving the S6K1‐dependent STAT3 phosphorylation and subsequently enhancing hepatic lipogenesis.

Novel approaches to oligonucleotide conjugation to a cell penetrating (Tat) peptide

open access: yes, 2009
Modifed oligonucleotides are routinely employed as bioanalytical probes for use in diagnostics; however, a major limiting factor in oligonucleotide-based diagnostics is poor cellular uptake.
