EIF4A3 enhances the viability, invasion and osteogenic differentiation of BMSCs via the USP53/SMAD5 pathway

open access: yesScientific Reports
SMAD5 has been demonstrated to promote osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs) through the circ_0001825/miR-1270/SMAD5 axis or KCNQ1OT1/miR-320a/SMAD5 axis.

Characteristics of Alveolar Bone Marrow Cells from Patients Undergoing Dental Extractions or Dental Implant Therapy [PDF]

open access: yes, 2020
Alveolar bone marrow stromal cells (aBMSCs) play important roles in craniofacial wound healing. To establish an easy, efficient and reliable method to harvest aBMSCs, we compared three different methods: extraction socket aspiration, osteotomy aspiration

Atrophic Skeletal Muscle‐Derived Extracellular Vesicles Transfer miR‐125a‐5p to Inhibit Bone Formation in Osteoporosis during Aging

open access: yesAdvanced Science, EarlyView.
A muscle‐bone endocrine pathway in aging is revealed in which extracellular vesicles released from atrophic skeletal muscle (Aged‐SKM‐EVs) inhibit bone formation. These EVs deliver miR‐125a‐5p to osteoblasts, thereby suppressing the SIRT7‐Sp7 signaling axis and osteogenic differentiation.

Targeting the GPX4–FUNDC1 Interaction with Magnesium Lithospermate B Attenuates Sepsis‐Associated Lung Injury

open access: yesAdvanced Science, EarlyView.
The diagram depicts the endothelial‐protective mechanism of magnesium lithospermate B (MLB) in sepsis‐associated lung injury. MLB binds GPX4 at Gly79, disrupts its interaction with FUNDC1, prevents mitophagy‐mediated GPX4 degradation, restores mitophagic flux, reduces ROS, and limits ferroptosis.

USP2-induced upregulation of LEF1 through deubiquitination relieves osteoporosis development by promoting the osteogenic differentiation of bone marrow mesenchymal stem cells

open access: yesJournal of Orthopaedic Surgery and Research
Background Bone marrow mesenchymal stem cells (BMSCs) exhibit therapeutic potential for osteoporosis through their differentiation into osteoblasts. Here, we investigated the role and mechanism of lymphoid enhancer-binding factor 1 (LEF1) in regulating ...

Developmentally Inspired, Mechanical–Metabolic Dual Gradient Osteochondral Constructs Bridging Regeneration and Therapeutic Screening

open access: yesAdvanced Science, EarlyView.
A developmentally inspired osteochondral construct integrates mechanical and metabolic dual gradients within a 3D‐printed platform. Human MSC spheroids are spatially directed to form cartilage–bone interfaces, enabling evaluation of regeneration without biochemical reagents and assessment of drug responses. This physiologically relevant, phase‐specific

Development of an early estimation method for predicting later osteogenic differentiation activity of rat mesenchymal stromal cells from their attachment areas

open access: yesScience and Technology of Advanced Materials, 2012
Cell morphology has received considerable attention in recent years owing to its possible relationship with cell functions, including proliferation, differentiation, and migration. Recent evidence suggests that extracellular environments can also mediate

Efficient generation of neural stem cell-like cells from adult human bone marrow stromal cells [PDF]

open access: yes, 2004
Clonogenic neural stem cells (NSCs) are self-renewing cells that maintain the capacity to differentiate into brain-specific cell types, and may also replace or repair diseased brain tissue. NSCs can be directly isolated from fetal or adult nervous tissue,

Single‐Cell Atlas of Subchondral Bone Marrow Lesions Reveals Proteostasis Dysfunction as a Druggable Mechanism for Early Osteoarthritis

open access: yesAdvanced Science, EarlyView.
This study reveals that abnormal mechanical stress downregulates the expression of HSP70 and impairs proteasome function in osteoarthritic bone cells, leading to misfolded collagen I accumulation and ER stress. When intracellular proteostasis capacity is exceeded, USP19 mediates the secretion of misfolded proteins into the extracellular space ...

Sirt3 Genetically Engineered Apoptotic Bodies Alleviate Skeletal Aging by Limiting Aggravated NLRP3 Inflammasome Activation of Senescent Macrophages

open access: yesAdvanced Science, EarlyView.
The senescence of macrophages leads to a decrease in the expression of Sirtuin3 (Sirt3), which in turn aggravates the NLRP3 inflammasome activation and IL‐1β secretion, thereby promoting the aging‐related osteoporosis. Sirt3‐enriched apoptotic bodies (ABs‐Sirt3) generated through genetic editing approaches can alleviate skeletal aging by targeting ...
