Identification of Novel Compounds Inhibiting the Kinase Activity of the CDK5/p25 Complex via Direct Binding to p25

Biochemistry, 2023
Tamoxifen, the gold standard drug for endocrine therapy for breast cancer, modulates the phosphorylation status of the TAU protein in Alzheimer's disease by inhibiting CDK5 kinase activity. Its binding to p25 prevents CDK5/p25 complexation and hence a decrease of CDK5 activity.

The P25 component of Galleria silk

Molecular and General Genetics MGG, 1998
The water-insoluble core of lepidopteran silk is composed of four major proteins, but only three genes have been identified. This study demonstrates that the 29- and 30-kDa components of Galleria mellonella silk are derived from a single gene designated P25.

Evaluation of the Interaction of Cyclin-Dependent Kinase 5 with Activator p25 and with p25-Derived Inhibitor CIP

Journal of Computational Biology, 2010
A high-affinity inhibitor protein called CIP, produced by small truncations of p35, was experimentally identified. P35 is a physiological activator of the cyclin-dependent kinase cdk5. P25 is derived from proteolytic truncation of p35 within "stressed" neurons, and it is associated with the hyperphosphorylation of specific neuronal proteins, typically ...

Controversies over p25 in Alzheimer's disease

Journal of Alzheimer’s Disease, 2002
Activity of the neuronal Cdc2-like kinase composed of cyclin-dependent kinase 5 (Cdk5) and its activator, p35, is critical for the normal development of the central nervous system, including cortical lamination and axonal patterning events. Conversely, altered Cdk5 activity has been associated with several neuronal defects including neurodegeneration ...

Proteolytic cleavage of Ii to p25.

The Journal of Immunology, 1988
Abstract The 25,000-Da protein that is seen in immunoprecipitates with antibodies to class II MHC molecules or to Ii was shown to be a C-terminal fragment of Ii. [35S]Methionine pulse-chase labeling of polyclonally activated B lymphocytes or lymphoblastoid cell lines demonstrated maximal appearance of p25 in Percoll-separated ...

Charge transport in dye-sensitized solar cells based on P25 TiO2 and amorphous-free P25 photoanodes

AIP Conference Proceedings, 2022
Previously we demonstrated that the elimination of the amorphous component in commercially available TiO2 (P25) has a favorable influence on performance of dye sensitized solar cells (DSCs). The photo-conversion efficiency (PCE) of DSCs based on hydrothermally treated, amorphous-free P25 (H-P25), sensitized with a commercial organometallic dye (N719 ...

Mechanism of CDK5/p25 Binding by CDK Inhibitors

Journal of Medicinal Chemistry, 2005
The cyclin-dependent kinases (CDK) CDK1, CDK2, CDK4, and CDK6 are serine/threonine protein kinases targeted in cancer therapy due to their role in cell cycle progression. The postmitotic CDK5 is involved in biological pathways important for neuronal migration and differentiation.

Philadelphia Chromosome with T(6;22) (P25;q12)

New England Journal of Medicine, 1976
The introduction of chromosome analysis in various blood disorders has proved the high specificity of the Philadelphia chromosome — a G22 member with deleted long arms — in the diagnosis of chronic...
