Cloning and functional characterisation of the mouse P2X7 receptor

open access: yesFEBS Letters, 1998
We have isolated a 1785-bp complementary DNA (cDNA) encoding the murine P2X7 receptor subunit from NTW8 mouse microglial cells. The encoded protein has 80% and 85% homology to the human and rat P2X7 subunits, respectively.

Mammalian P2X7 receptor pharmacology: comparison of recombinant mouse, rat and human P2X7 receptors [PDF]

open access: yesBritish Journal of Pharmacology, 2009
Background and purpose: Acute activation of P2X7 receptors rapidly opens a non‐selective cation channel. Sustained P2X7 receptor activation leads to the formation of cytolytic pores, mediated by downstream recruitment of hemichannels to the cell surface.

P2X7 receptors in the central nervous system

Biochemical Pharmacology, 2021
For the past three decades, our laboratory has conducted pioneering research to elucidate the complexity of purinergic signaling in the CNS, alone and in collaboration with other groups, inspired by the ground-breaking efforts of Geoffrey Burnstock. This review summarizes our contribution to understand the nucleotide receptor signaling in the CNS with ...

Novel P2X7 receptor antagonists

Bioorganic & Medicinal Chemistry Letters, 2003
The synthesis and pharmacological evaluation of a new series of potent P2X(7) receptor antagonists is disclosed. The compounds inhibit BzATP-mediated pore formation in THP-1 cells. The distribution of the P2X(7) receptor in inflammatory cells, most notably the macrophage, mast cell and lymphocyte, suggests that P2X(7) antagonists have a significant ...

P2X7 receptors in the nervous system

Progress in Neurobiology, 2006
P2X(7) receptors are ligand-gated ion channels, expressed as homo-oligomeric assemblies of individual subunits. They are widely distributed at immunocompetent cells of the central and peripheral nervous system and are believed to be primarily involved in host-defense reaction. However, a growing amount of evidence indicates that their signaling role in
