Research Progress of Pectin Methylesterase and its Inhibitors

Current Protein and Peptide Science, 2022
Abstract:As an important pectin enzyme, pectin methylesterase (PME) can hydrolyze methyl esters, release methanol and reduce esterification. It is essential in regulating pollen tube development, root extension, and fruit ripening. Pectin methylesterase inhibitors (PMEI) can specifically bind PME and inhibit its activity, which jointly determines the ...

Aspergillus aculeatus pectin methylesterase: study of the inactivation by temperature and pressure and the inhibition by pectin methylesterase inhibitor

Enzyme and Microbial Technology, 2005
Abstract The thermal inactivation kinetics of purified Aspergillus aculeatus pectin methylesterase (PME) was investigated. In the temperature range 46–56 °C A. aculeatus PME inactivates following a first-order reaction. Addition of CaCl 2 (50 mM or 1.0 M) has no noticeable influence on the inactivation parameters. A.

Pectin methylesterase and polygalacturonase in the developing grape skin

Plant Physiology and Biochemistry, 2008
Ripening of grape (Vitis vinifera L.) berry immediately precedes harvesting and the evolution of the skin tissue is important as it contains the key compounds for wine quality. Grape softening is thought to result from extensive cell wall modifications that occur during ripening.

Pectin methylesterase and its proteinaceous inhibitor: a review

Carbohydrate Research, 2010
Pectin methylesterase (PME) catalyses the demethoxylation of pectin, a major plant cell wall polysaccharide. Through modification of the number and distribution of methyl-esters on the pectin backbone, PME affects the susceptibility of pectin towards subsequent (non-) enzymatic conversion reactions (e.g., pectin depolymerisation) and gel formation, and,

Activity of an atypical Arabidopsis thaliana pectin methylesterase

Planta, 2008
An Arabidopsis thaliana pectin methylesterase that was not predicted to contain any signaling sequence was produced in E. coli and purified using a His tag added at its N-terminus. The enzyme demethylesterified Citrus pectin with a Km of 0.86 mg/ml. The enzyme did not require salt for activity and was found to be relatively temperature-sensitive.

Pectin methylesterase inhibitor

Biochimica et Biophysica Acta (BBA) - Proteins and Proteomics, 2004
Pectin methylesterase (PME) is the first enzyme acting on pectin, a major component of plant cell wall. PME action produces pectin with different structural and functional properties, having an important role in plant physiology. Regulation of plant PME activity is obtained by the differential expression of several isoforms in different tissues and ...

Characterization of carrot pectin methylesterase

Cellular and Molecular Life Sciences (CMLS), 2002
The most alkaline form of pectin methylesterase was purified from ripe carrot roots and used for structural analysis. Determination of an N-terminal blocking group and of the primary structure allowed comparisons with other forms, and facilitated crystallographic determination of the three-dimensional structure.

A Continuous Fluorometric Assay for Pectin Methylesterase

Analytical Biochemistry, 1996
A continuous, fluorometric assay for pectin methylesterase (PME) activity is described. In this assay, methanol produced by PME hydrolysis of pectin methyl esters is oxidized to formaldehyde by alcohol oxidase, and the formaldehyde is continuously reacted with 4-amino-3-penten-2-one to create a stable, fluorescent product.

Inhibition of tomato pectin methylesterase by partially purified kiwi pectin methylesterase inhibitor protein

International Journal of Food Science & Technology, 1994
The inhibition of tomato pectin methylesterase (PME) by a recently discovered kiwi pectin methylesterase inhibitor (PMEI) is described. PME was consequently purified by CM Sephadex C‐50, Concanavalin A‐Sepharose 4 B and Mono S chromatography, and PMEI by Q‐Sepharose and Sephacryl S‐200 chromatography.
