Multi‐tissue Metabolic GWAS and Drought‐Responsive Multi‐omics Reveal the Genetic Basis of the Quinoa Metabolome

open access: yesAdvanced Science, EarlyView.
A multi‐omics framework combining multitissue genome‐wide association studies, metabolomics, transcriptomics, proteomics, and functional validation uncovers the genetic basis of specialized metabolism in quinoa. The study identifies hundreds of metabolite‐associated loci, prioritizes candidate genes for saponin, betalain, and flavonoid biosynthesis ...

LRRC4 Orchestrates AP2A1‐Containing Clathrin‐Coated Vesicles to Disrupt Mitochondrial Cristae and Restrict Glioblastoma Progression

open access: yesAdvanced Science, EarlyView.
LRRC4 suppresses GBM by regulating AP2A1‐containing Golgi‐derived clathrin‐coated vesicles. Low LRRC4 allows cytoplasmic AP2A1 to maintain mitochondrial fission‐fusion, MICOS integrity, and OXPHOS, promoting proliferation and invasion. High LRRC4 redirects AP2A1‐containing Golgi‐derived clathrin‐coated vesicles to mitochondria, disrupts MICOS, enhances

PEAR1 Promotes Glucose Metabolism Reprogramming in Sepsis‐Associated Acute Lung Injury via AARS1‐Mediated HIF‐1α Lactylation

open access: yesAdvanced Science, EarlyView.
This study revealed that a PEAR1/HIF‐1α/ glycolysis/lactate/H3K18la positive feedback loop in PMVECs that drives the development of S‐ALI. Mechanistically, PEAR1 mediates the binding of HIF‐1α to AARS1, leading to the lactylation of HIF‐1α, the primary lactylation site of which is K172.

Draft genome sequences of Cylindrospermopsis raciborskii strains CS-508 and MVCC14, isolated from freshwater bloom events in Australia and Uruguay

open access: yesStandards in Genomic Sciences, 2018
Members of the genus Cylindrospermopsis represent an important environmental and health concern. Strains CS-508 and MVCC14 of C. raciborskii were isolated from freshwater reservoirs located in Australia and Uruguay, respectively.

Synthesis and characterization of a specific peptide nucleic acid that inhibits expression of inducible NO synthase

open access: yesFEBS Letters, 1998
Inducible nitric oxide synthase (iNOS) is modulated at the transcriptional level. Overexpression of this protein may result in high levels of nitric oxide leading to tissue damage and immunosuppression. In order to reduce the pathological effects of NO overproduction many efforts have been devoted to the identification of specific inhibitors of iNOS ...

Microprotein MP104 Promotes Malignant Progression of Colorectal Cancer Through Regulating Protein Translation

open access: yesAdvanced Science, EarlyView.
A previously unrecognized microprotein, MP104, encoded by ZEB1‐AS1, emerges as a critical driver of colorectal cancer progression. MP104 links ubiquitin‐mediated protein degradation with translational reprogramming via the UBE2O–AMPKα2–mTOR–EIF4B axis, revealing an unrecognized layer of oncogenic regulation.

A Wagner–Meerwein‐Like Rearrangement Generates a Vinyl Group in the Biosynthesis of the Polychlorinated Lipopeptides Fischerazoles

open access: yesAngewandte Chemie, EarlyView.
The fischerazoles, unusual cyanobacterial chlorinated lipopeptides are reported. During their biosynthesis, a linear fatty acyl‐acyl carrier protein (ACP) precursor is rearranged through the action of the S‐adenosyl‐methionine (SAM)‐dependent methyltransferase FshF, to generate a pendant vinyl group.

Crystal structure of the condensation domain from lovastatin polyketide synthase

open access: yesSynthetic and Systems Biotechnology, 2019
The highly reducing iterative polyketide synthases responsible for lovastatin biosynthesis contains a section homologous to condensation (CON) domain observed in nonribosomal peptide synthetases (NRPSs).

Pyrazole‐Derived Antibacterial Compounds Effectively Treat Methicillin‐Resistant Staphylococcus Aureus Infections by Inhibiting Aspartate Transcarbamoylase

open access: yesAdvanced Science, EarlyView.
This study unveils a novel antibacterial mechanism against MRSA, in which the compound Py‐27 selectively targets and inhibits aspartate transcarbamoylase (ATCase), a key enzyme in pyrimidine synthesis. This inhibition disrupts DNA replication, induces oxidative damage, leading to potent bactericidal activity.
