An inframe perforin gene deletion in familial hemophagocytic lymphohistiocytosis is associated with perforin expression [PDF]
Familial hemophagocytic lymphohistiocytosis is an autosomal recessive disease of early childhood manifested by hypercytokinemia and organ infiltration of macrophages and activated lymphocytes, and it is characterized by a fulminant clinical course. The molecular mechanism underlying this disease appears to be a deregulation of apoptosis of activated T ...

PLD3 activates the lysosomal‐AKT‐NF‐κB axis to drive cellular senescence in macrophages, establishing an immunosuppressive TME by limiting the infiltration of cytotoxic T, NK, and NKT cells, which confers resistance to anti‐PD‐1 therapy. Abrine inhibits PLD3 expression, restoring antitumor immunity and synergizing with anti‐PD‐1 treatment.

PGK1 in tumor cells upregulates CCL2 expression through activation of the AKT/GSK‐3β/β‐catenin signaling axis, thereby promoting the recruitment and M2 polarization of TAMs and ultimately impairing the infiltration and activation of CD8+ T cells within the HCC tumor microenvironment. ABSTRACT Patients with advanced hepatocellular carcinoma (HCC) have a

This study develops 16:0 LPC‐modified lipid nanoparticles (LPC‐LNPs) with cancer cell specificity by exploiting altered tumor lipid metabolism. LPC‐LNPs encapsulating Cd28 small interfering RNA (LPC‐LNP‐Cd28) knock down cancer cell CD28 without affecting T cells, inflame the tumor microenvironment, and overcome anti‐PD‐1 resistance.

Single‐cell RNA editing analysis identifies ADAR1 as a regulator of dysfunctional T cell states in colorectal cancer. Elevated ADAR1 activity promotes T cell exhaustion and impairs antitumor immunity partly through TGF‐β‐SMAD signaling, contributing to anti‐PD‐1 resistance and highlighting T cell ADAR1 as a potential therapeutic target and biomarker ...

The Role of Perforin in Infections and Tumour Surveillance [PDF]
Cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells are able to lyse suitable target cells. CTLs recognize a specific peptide epitope presented by major histocompatibility complex (MHC) class I molecules on the target cells, whereas NK cells lyse targets that express no or low MHC class I molecules.

UBE2O targets CDKL1 for the degradation to upregulate PD‐L1 expression and promotes resistance to radioimmunotherapy in lung cancer, supporting the potential of targeting UBE2O as a promising therapeutic strategy. ABSTRACT Resistance to radioimmunotherapy is one of the primary causes of treatment failure in lung cancer patients; however, the underlying

Immunological profile and its clinical implications in pediatric infectious mononucleosis
ObjectiveTo analyze the differences in peripheral blood immune function between children with infectious mononucleosis (IM) who developed liver injury (LI) and those non-liver injury (NLI), and to investigate the regulatory role of immune markers in IM ...
