Phosphatidylinositol-4-phosphate controls autophagosome formation in Arabidopsis thaliana [PDF]

open access: yesNature Communications, 2022
Autophagosomes are specialized vesicles that target and deliver cargo to the lytic vacuole. Here the authors show that plasma-membrane derived lipid phosphatidylinositol-4-phosphate supports the assembly and expansion of autophagosomes in ...

OSBP is a Major Determinant of Golgi Phosphatidylinositol 4-Phosphate Homeostasis [PDF]

open access: yesContact, 2023
The lipid phosphatidylinositol 4-phosphate (PI4P) plays a master regulatory role at Golgi membranes, orchestrating membrane budding, non-vesicular lipid transport and membrane organization.

Plasma Membrane Phosphatidylinositol 4-Phosphate Is Necessary for Virulence of Candida albicans [PDF]

open access: yesmBio, 2022
Phosphatidylinositol lipids regulate key processes, including vesicle trafficking and cell polarity. A recent study identified novel roles for phosphatidylinositol 4-phosphate (PI4P) in the plasma membrane of the fungal pathogen Candida albicans ...

SAC1 regulates autophagosomal phosphatidylinositol-4-phosphate for xenophagy-directed bacterial clearance [PDF]

open access: yesCell Reports, 2021
Summary: Phosphoinositides are important molecules in lipid signaling, membrane identity, and trafficking that are spatiotemporally controlled by factors from both mammalian cells and intracellular pathogens.

Membrane pools of phosphatidylinositol-4-phosphate regulate KCNQ1/KCNE1 membrane expression [PDF]

open access: yesCommunications Biology, 2021
Braun et al. investigate the effects of PI 4-kinases and their lipid product, PI4P, in KCNQ channel function and sub-cellular localization. These results suggest a possible harmful effect of PI4K inhibition on cardiac electrophysiology and suggest the ...

An acquired phosphatidylinositol 4-phosphate transport initiates T-cell deterioration and leukemogenesis [PDF]

open access: yesNature Communications, 2022
The oxysterol-binding protein-related protein 4 (ORP4L) is expressed in T-cell acute lymphoblastic leukemia and is required for leukemogenesis. Here the authors show that ORP4L orchestrates the transport of the phospholipid PI(4)P from Golgi to the ...

Phosphatidylinositol-4-phosphate joins the dance of plant autophagosome formation. [PDF]

open access: yesAutophagy, 2023
In plants, macroautophagy/autophagy is a key mechanism that contributes to their ability to cope with a wide range of environmental constraints such as drought, nutrient starvation or pathogen resistance. Nevertheless, the molecular mechanisms of plant autophagy, and notably that of autophagosome formation, remain poorly understood.

Multiphasic dynamics of phosphatidylinositol 4-phosphate during phagocytosis. [PDF]

open access: yesMol Biol Cell, 2017
We analyzed the distribution, fate, and functional role of phosphatidylinositol 4-phosphate (PtdIns4P) during phagosome formation and maturation. To this end, we used genetically encoded probes consisting of the PtdIns4P-binding domain of the bacterial effector SidM. PtdIns4P was found to undergo complex, multiphasic changes during phagocytosis.

Phosphatidylinositol 4-phosphate and phosphatidylinositol 3-phosphate regulate phagolysosome biogenesis. [PDF]

open access: yesProc Natl Acad Sci U S A, 2015
Significance Phagocytes are key to innate immunity, as they ingest, kill, and degrade foreign particles, such as microbes. Particle uptake results in the formation of phagosomes that eventually fuse with lysosomes, the main degradative compartments in cells.

Phosphatidylinositol 4-phosphate 5-kinases in the regulation of T cell activation [PDF]

open access: yesFrontiers in Immunology, 2016
Phosphatidylinositol 4,5-biphosphate kinases (PIP5K) are critical regulators of T cell activation being the main enzymes involved in the synthesis of phosphatidylinositol 4,5-biphosphate (PIP2).
