Justification of widened dissolution specifications of an extended-release product using physiologically based biopharmaceutics modeling

open access: yesXenobiotica
Drug products meeting the dissolution specifications is crucial in order to ensure consistent clinical performance. However, in certain cases, wider dissolution specifications may be required based on product behaviour. While the justification of such wider specifications may be challenging from a regulatory context, approaches such as physiological ...

Structural Insights Into the Function of Leishmania major Adenylosuccinate Lyase

open access: yesProteins: Structure, Function, and Bioinformatics, EarlyView.
ABSTRACT One of several intriguing aspects of kinetoplastid biochemistry is the complete dependence on host purines and purine recycling due to the lack of a de novo purine biosynthesis pathway. Adenylosuccinate lyase (ASL, EC 4.3.2.2) is a key enzyme in the purine synthesis pathway responsible for the conversion of adenylosuccinate into adenosine ...

Advancing pharmacometrics in Africa—Transition from capacity development toward job creation

open access: yesCPT: Pharmacometrics &Systems Pharmacology, Volume 14, Issue 3, Page 407-419, March 2025.
Abstract Trained pharmacometricians remain scarce in Africa due to limited training opportunities, lack of a pharmaceutical product development ecosystem, and emigration to high‐income countries. The Applied Pharmacometrics Training (APT) fellowship program was established to address these gaps and specifically foster job creation for talent retention.

Thermosensitive Nanostructured Coacervates Developed by Mixing ABA‐ and AB‐Block Copolymers and Their Unique Sustained Release of Nano‐Micelles in a Regulated Manner

open access: yesSmall, EarlyView.
Polyion complex micelles composed of blends of ABA and AB block copolymers can form coacervates with a hexagonally packed cylinder nanostructure after heating. The resulting coacervates can release the micelles under physiological conditions in a zero‐order kinetics, possessing cargo proteins.
