Structural biology of ferritin nanocages

open access: yesFEBS Letters, EarlyView.
Ferritin is a conserved iron‐storage protein that sequesters iron as a ferric mineral core within a nanocage, protecting cells from oxidative damage and maintaining iron homeostasis. This review discusses ferritin biology, structure, and function, and highlights recent cryo‐EM studies revealing mechanisms of ferritinophagy, cellular iron uptake, and ...

In vitro models of cancer‐associated fibroblast heterogeneity uncover subtype‐specific effects of CRISPR perturbations

open access: yesMolecular Oncology, EarlyView.
Development of therapies targeting cancer‐associated fibroblasts (CAFs) necessitates preclinical model systems that faithfully represent CAF–tumor biology. We established an in vitro coculture system of patient‐derived pancreatic CAFs and tumor cell lines and demonstrated its recapitulation of primary CAF–tumor biology with single‐cell transcriptomics ...

Selection of Reference Genes by Quantitative Real-Time PCR in Different Cell Lines from Humpback Grouper (Cromileptes altivelis)

open access: yesFishes
Humpback grouper (Cromileptes altivelis) is an economically important fish, but the increasing density of its farming has led to more severe disease outbreaks.

ATF4‐mediated stress response as a therapeutic vulnerability in chordoma

open access: yesMolecular Oncology, EarlyView.
We screened 5 chordoma cell lines against 100+ inhibitors of epigenetic and metabolic pathways and kinases and identified halofuginone, a tRNA synthetase inhibitor. Mechanistically halofuginone induces an integrated stress response, with eIF2alpha phosphorylation, activation of ATF4 and its target genes CHOP, ASNS, INHBE leading to cell death ...

High Yielding, One-Pot Synthesis of Bis(1H-indazol-1-yl)methane Catalyzed by 3d-Metal Salts

open access: yesReactions, 2022
Synthetic access to poly(indazolyl)methanes has limited their study despite their structural similarity to the highly investigated chelating poly(pyrazolyl)methanes and their potentially important indazole moiety. Herein is presented a high yielding, one-

Ivermectin improved behavioral and molecular deficits of poly I: C-Induced schizophrenia model in rats

open access: yesNeuroscience Applied, 2023
Backrounds: Schizophrenia is a devastating psychiatric disorder with complex symptoms and neurobiology, affecting approximately 1% of the world's population. Studies have shown that exposure to viral and bacterial infections during pregnancy increases the risk of neurodevelopmental disorders including schizophrenia and autism  ...

PARP inhibition and pharmacological ascorbate demonstrate synergy in castration‐resistant prostate cancer

open access: yesMolecular Oncology, EarlyView.
Pharmacologic ascorbate (vitamin C) increases ROS, disrupts cellular metabolism, and induces DNA damage in CRPC cells. These effects sensitize tumors to PARP inhibition, producing synergistic growth suppression with olaparib in vitro and significantly delayed tumor progression in vivo. Pyruvate rescue confirms ROS‐dependent activity.

Toll-like receptor activation induces airway obstruction and hyperresponsiveness in guinea pigs

open access: yesRespiratory Research
Background Microbial infections, particularly those caused by rhinovirus (RV) and respiratory syncytial virus (RSV), are major triggers for asthma exacerbations. These viruses activate toll-like receptors (TLRs), initiating an innate immune response.

[Interferon induction in monkeys by poly I-C complex with poly-L-lysine].

open access: yesVoprosy virusologii, 1977
The spectrum characteristics, the stability to pancreatic RN-ase and hydrolases of the monkey and mouse blood as well as interferon induction in mice and monkeys by poly I: poly C and its complex with poly-l-lysine were studied. The complex was found to be more stable to the treatments tested as well as markedly interferonogenically active in Macaca ...

Plecstatin inhibits hepatocellular carcinoma tumorigenesis and invasion through cytolinker plectin

open access: yesMolecular Oncology, EarlyView.
The ruthenium‐based metallodrug plecstatin exerts its anticancer effect in hepatocellular carcinoma (HCC) primarily through selective targeting of plectin. By disrupting plectin‐mediated cytoskeletal organization, plecstatin inhibits anchorage‐dependent growth, cell polarization, and tumor cell dissemination.
