Harnessing Microbial Interactions to Elicit the Production of Polyene Macrolides in Streptomyces [PDF]
Since their discovery in the 1950s, polyene macrolides have been invaluable for treating life threatening fungal infections. Polyene antibiotics are unique because they target ergosterol, the fungal sterol, and disrupt the cellular processes that rely on

Engineered biosynthesis and characterization of disaccharide-pimaricin
Background Disaccharide polyene macrolides exhibit superior water solubility and significantly reduced hemolytic toxicity compared to their monosaccharide counterparts, making them promising candidates for safer antifungal therapeutics. In this study, we

Potentiation of antiviral activity of acyclovir by polyene macrolide antibiotics [PDF]
The potentiation of the antiviral activity of acyclovir [9-[(2-hydroxyethoxy)methyl]guanine] by polyene macrolide antibiotics has been studied as a function of the macrolide structure. The 12 polyenes chosen for this study represented the major structural groups of these antibiotics and induced in mammalian cells repairable membrane alterations or ...

Many antibiotics originate from soil‐inhabiting Actinobacteria, especially from the diverse genus Streptomyces. However, the emergence of antibiotic resistance poses a significant global challenge to treating infectious diseases. Therefore, the search for Actinobacteria, particularly from less‐explored environments, as potential sources of novel ...

Modular Synthesis of Dihydropyranones through Cyclization and Organometallic Functionalization
A scalable method for synthesizing 3‐methoxy‐4,5‐dihydropyran‐2‐ones (MeO‐DHP‐2‐ones) via base‐promoted cyclization of hydroxy alkynes is presented. Mechanistic and computational studies guided the transformation into 4,5‐dihydropyran‐4‐ones (DHP‐4‐ones), enabling access to spiroacetal precursors relevant to natural product synthesis.
