Harnessing Microbial Interactions to Elicit the Production of Polyene Macrolides in Streptomyces [PDF]

open access: yes, 2018
Since their discovery in the 1950s, polyene macrolides have been invaluable for treating life threatening fungal infections. Polyene antibiotics are unique because they target ergosterol, the fungal sterol, and disrupt the cellular processes that rely on

Engineered biosynthesis and characterization of disaccharide-pimaricin

open access: yesMicrobial Cell Factories
Background Disaccharide polyene macrolides exhibit superior water solubility and significantly reduced hemolytic toxicity compared to their monosaccharide counterparts, making them promising candidates for safer antifungal therapeutics. In this study, we

Potentiation of antiviral activity of acyclovir by polyene macrolide antibiotics [PDF]

open access: yesAntimicrobial Agents and Chemotherapy, 1984
The potentiation of the antiviral activity of acyclovir [9-[(2-hydroxyethoxy)methyl]guanine] by polyene macrolide antibiotics has been studied as a function of the macrolide structure. The 12 polyenes chosen for this study represented the major structural groups of these antibiotics and induced in mammalian cells repairable membrane alterations or ...

Significant Improvement of Antifungal Activity of Polyene Macrolides by Bisalkylation of the Mycosamine

open access: yes, 2016
New derivatives of Amphotericin B (AmB) were synthesized through a double reductive alkylation of the mycosamine. These derivatives of AmB displayed superior antifungal activity against Saccharomyces cerevisiae wild-type strain and especially in the case

Screening the Antibiotic Activity of Cave Actinobacteria Against Multidrug‐Resistant Strains of Pseudomonas aeruginosa and Methicillin‐Resistant Staphylococcus aureus

open access: yesInternational Journal of Microbiology, Volume 2026, Issue 1, 2026.
Many antibiotics originate from soil‐inhabiting Actinobacteria, especially from the diverse genus Streptomyces. However, the emergence of antibiotic resistance poses a significant global challenge to treating infectious diseases. Therefore, the search for Actinobacteria, particularly from less‐explored environments, as potential sources of novel ...

Probing the Structure−Function Relationship of Polyene Macrolides:  Engineered Biosynthesis of Soluble Nystatin Analogues

open access: yes, 2016
Although polyene macrolides are efficient antifungal agents with fungicidal mode of action, their use in medical practice is problematic due to their low solubility and significant human toxicity.

Digitoxose as powerful glycosyls for building multifarious glycoconjugates of natural products and un-natural products

open access: yesSynthetic and Systems Biotechnology
Digitoxose, a significant 2,6-dideoxyhexose found in nature, exists in many small-molecule natural products. These digitoxose-containing natural products can be divided into steroids, macrolides, macrolactams, anthracyclines, quinones, enediynes, acyclic

Modular Synthesis of Dihydropyranones through Cyclization and Organometallic Functionalization

open access: yesEuropean Journal of Organic Chemistry, Volume 28, Issue 48, December 22, 2025.
A scalable method for synthesizing 3‐methoxy‐4,5‐dihydropyran‐2‐ones (MeO‐DHP‐2‐ones) via base‐promoted cyclization of hydroxy alkynes is presented. Mechanistic and computational studies guided the transformation into 4,5‐dihydropyran‐4‐ones (DHP‐4‐ones), enabling access to spiroacetal precursors relevant to natural product synthesis.
