Potato is widely consumed across the globe. Understanding and inhibiting the oxidation caused by polyphenol oxidase (PPO) could improve shelf life and increase the nutritional and economic value of potato proteins. This study aimed to identify and quantify all expressed PPOs in potato tuber (Solanum tuberosum) and to purify and characterize the major ...

Abstract Recent progress in the study of plant polyphenol oxidases is critically reviewed. Two main groups are recognized: the catecholoxidases and the laccases. Their purification, subcellular location and protein properties are described. Attention is also given to their activation and induction, their function and evolution.

The enzyme polyphenol oxidase (PPO) has been implicated in discoloration of Asian noodles. The recombinant inbred line (RIL) populations, M6/‘Opata 85’, NY18/CC, and ND2603/‘Butte 86’ were used to investigate the distribution, chromosome location, and number of loci involved in wheat (Triticum aestivum L.) PPO. PPO activity was measured by means of the

Polyphenol Oxidase as a Factor in Aging
Gerontology, 2009Polyphenol oxidase oxidizes several proteins that contain tyrosine. During aging, when synthesis and replacement of catabolized proteins are lagging, the relative proportion of oxidized species might reach high values, thus impairing physiological function. A mathematical analysis verifies this assumption.

Polyphenol Oxidase‐Like Nanozymes
Advanced MaterialsAbstract Nanozymes are nanomaterials with enzyme‐like activities. Over the past decade, a steadily growing body of research on polyphenol oxidase‐like (PPO‐like) nanozymes, a subgroup of oxidase‐like nanozymes, has suggested their increasing prominence in nanozymology. Most importantly, PPO‐like nanozymes have developed a unique, well‐

Recombinant polyphenol oxidases for production of theaflavins from tea polyphenols
International Journal of Biological Macromolecules, 2019Theaflavins (TFs) have attracted much attention due to their various bioactivities in black tea. This paper describes the first trial for enzymatic production of TFs by recombinant polyphenol oxidases (PPOs). PPO genes were cloned from nine species and expressed in E. coli.

Inhibition of Mushroom Polyphenol Oxidase by Agaritine
Journal of Agricultural and Food Chemistry, 1998Agaritine [(β-N-(γ-l(+)-glutamyl)-4-hydroxymethylphenylhydrazine] was isolated and purified from mushrooms (Agaricus bisporus). This paper reports for the first time the inhibition of the monophenolase and diphenolase activities of mushroom polyphenol oxidase by this abundant and characteristic compound from the Agaricus genus.

Oxidation of the flavonol fisetin by polyphenol oxidase
Biochimica et Biophysica Acta (BBA) - General Subjects, 1998The present study demonstrates the antiradical efficiency of fisetin, a flavonol widely distributed in fruits and vegetables, by its ability to react with two different free radicals, ABTS; and DPPH;. The polyphenolic nature of fisetin led us to consider whether it might be oxidised by polyphenol oxidase (PPO), and the results reported show that it can

Properties of tobacco polyphenol oxidase
Archives of Biochemistry and Biophysics, 1959Abstract The properties of partially purified tobacco leaf polyphenol oxidase have been studied. Evidence is presented to establish that the observed oxidations of catechol, caffeic acid, and chlorogenic acid were, in fact, catalyzed by polyphenol oxidase. Enzyme stability, activation, x-ray inactivation, inhibition, pH optima, substrate specificity,

The Purification of Polyphenol Oxidase from Tobacco
Protein Expression and Purification, 2002A new polyphenol oxidase (PPO) named PPO II was purified from tobacco (Nicotiana tobacum) by using acetone powder, ammonium sulfate precipitation, and column chromatography on DEAE-Sephadex A-50, Sephadex G-75, and CM-Sephadex C-50. It has an active site of a pair of type 3 coppers bridged to phenolate oxygen, which represents a new catalytic mechanism
