GPCRs in CAR‐T Cell Immunotherapy: Expanding the Target Landscape and Enhancing Therapeutic Efficacy

open access: yesAdvanced Science, EarlyView.
Chimeric antigen receptor T cell therapy faces dual challenges of target scarcity and an immunosuppressive microenvironment in solid tumors. This review highlights how G protein‐coupled receptors can serve as both novel targets to expand the therapeutic scope and functional modules to enhance CAR‐T cell efficacy.

Genetic deletion of neuronal PPARγ enhances the emotional response to acute stress and exacerbates anxiety: An effect reversed by rescue of amygdala PPARγ function [PDF]

open access: yes, 2016
PPARγ is one of the three isoforms of the Peroxisome Proliferator-Activated Receptors (PPARs). PPARγ is activated by thiazolidinediones such as pioglitazone, and it is targeted to treat insulin resistance.

Curcumin Attenuates Asthmatic Airway Inflammation and Mucus Hypersecretion Involving a PPARγ-Dependent NF-κB Signaling Pathway In Vivo and In Vitro

open access: yesMediators of Inflammation, 2019
Asthma is characterized by airway inflammation and mucus hypersecretion. Curcumin possessed a potent anti-inflammatory property involved in the PPARγ-dependent NF-κB signaling pathway.

A Simplified Three‐Tailed N‐Alkyl Phosphoramidate Lipid Platform Enables Inguinal Adipose‐Accumulated mRNA Delivery for Anti‐Obesity Therapy

open access: yesAdvanced Science, EarlyView.
This study develops an mRNA therapy encoding a long‐acting GLP‐1/FGF21 fusion protein. A three‐tailed ionizable lipid engineered three‐component lipid nanoparticle delivers the mRNA to inguinal adipose tissue. In obese mice, this targeted, dual‐hormone treatment potently reduces body weight, fat mass, presenting a localized strategy for metabolic ...

The anti-atherogenic effects of thiazolidinediones [PDF]

open access: yes, 2007
The thiazolidinediones (TZDs) rosiglitazone (ROS) and pioglitazone (PIO) are insulin-sensitising agents widely used to treat patients with type 2 diabetes mellitus (T2DM). Thiazolidinediones significantly improve glycaemic control in diabetics by reduced

Collaborative Power of Nrf2 and PPARγ Activators against Metabolic and Drug-Induced Oxidative Injury

open access: yesOxidative Medicine and Cellular Longevity, 2017
Mammalian cells have evolved a unique strategy to protect themselves against oxidative damage induced by reactive oxygen species (ROS). Especially, two transcription factors, nuclear factor erythroid 2p45-related factor 2 (Nrf2) and peroxisome ...

Colorectal Cancer Cell's Weapon: RNF32 Engages SPP1+ Macrophages to Foster Liver Metastasis, Targeted by Indole‐3‐Acetic Acid

open access: yesAdvanced Science, EarlyView.
Cytoplasmic RNF32 fuels CRC liver metastasis by degrading GSK3β, which stabilizes β‐catenin and activates Wnt/EMT. Moreover, RNF32 rewires the metastatic niche: it depletes CD8+/CD4+ T and NK cells while recruiting SPP1+ macrophages (which boost tumor stemness via CD44), fibroblasts, and immunosuppressive monocytes to aid colonization.

Structural Basis for the Regulation of PPARγ Activity by Imatinib

open access: yesMolecules, 2019
Imatinib is an effective anticancer drug for the treatment of leukemia. Interestingly, when an FDA-approved drug library was tested for agents that block peroxisome proliferator-activated receptor γ (PPARγ) phosphorylation at Ser245 to evaluate

Potential use of human periapical cyst-mesenchymal stem cells (hPCy-MSCs) as a novel stem cell source for regenerative medicine applications [PDF]

open access: yes, 2017
Mesenchymal stem cells (MSCs) are attracting growing interest by the scientific community due to their huge regenerative potential. Thus, the plasticity of MSCs strongly suggests the utilization of these cells for regenerative medicine applications.

PPARγ is a nexus controlling alternative activation of macrophages via glutamine metabolism

open access: yesGenes & Development, 2018
Nelson et al. report that peritoneal macrophage respiration is enhanced by rosiglitazone, an activating PPARγ ligand, in a PPARγ-dependent manner.
