Functional evolution of the pregnane X receptor [PDF]
The pregnane X receptor (PXR; NR1I2) is a nuclear hormone receptor (NR) that transcriptionally regulates genes encoding transporters and drug-metabolising enzymes in the liver and intestine. PXR activation leads to enhanced metabolism and elimination of xenobiotics and endogenous compounds such as hormones and bile salts.

Synthesis of 6,19-Sulfamidate Bridged Pregnanes
Conformationally restrained substituted pregnane-20-one derivatives were obtained by an intramolecular nitrene addition onto a C-5/C-6 double bond involving a tethered C-19 sulfamoyl moiety. The resulting aziridine underwent regioselective nucleophilic ring opening at C-5 at room temperature with cyanide, fluoride, and acetate.

Ring a Aromatic Steroids in the Pregnane Series
4-Methylpregna-1,3,5(10)-trien-20-one and 4-methylpregna-1,3,5(10),16-tetraen-20-one have been obtained by a dienol: benzene type of rearrangement of 5α,6α-epoxypregnanes possessing a further double bond equivalent on ring A. The crystal structure of 4-methyl-19-norpregna-1,3,5(10)-trien-20-one is reported.

Objective Members of the superfamily of nuclear receptors have been implicated in inflammatory processes and pathologic tissue remodeling and have emerged as attractive targets for pharmaceutical intervention. However, the role of testicular receptor 4 (TR4; or Nr2c2) in fibroblast activation and rheumatologic diseases has not yet been investigated ...
