We present a pathology‐responsive, activity‐based prodrug strategy that tackles multiple hallmarks of Alzheimer's disease (AD). Rationally designed BE compounds convert under oxidative stress into redox‐active aminophenols, neutralizing ROS and redirecting both metal‐free and metal‐bound Aβ aggregation.

This study presents a novel glutathione‐activatable prodrug nanomedicine (cRGD‐PEG‐PGlu‐NP (ssTrypsin & CDDP)) that co‐encapsulates cisplatin and a masked protease to combat cisplatin resistance. The dual‐drug system selectively targets resistant tumor cells, enhancing the efficacy of cisplatin while minimizing systemic toxicity.
