Expression, purification and characterization of the human proton-coupled folate transporter [PDF]

open access: yes, 2016
The human proton-coupled folated transporter (PCFT, SLC46A1) is the primary means for the absorption of dietary folates. Folates (Vitamin B9) and folate cofactors are required for more than a hundred metabolic reactions in cells including synthesis of ...

Site-specific contribution of proton-coupled folate transporter/haem carrier protein 1 in the intestinal absorption of methotrexate in rats

open access: yes, 2009
Objectives Methotrexate is reportedly a substrate for proton-coupled folate transporter/haem carrier protein 1 (PCFT/HCP1) and reduced folate carrier 1 (RFC1).

Structural Characterization And Therapeutic Utility Of The Proton-Coupled Folate Transporter [PDF]

open access: yes, 2016
Folate is a B9 vitamin essential to DNA synthesis. The proton-coupled folate transporter (PCFT) is a newly discovered proton/folate symporter with an acidic pH optimum and broad expression across a variety of solid tumor types, with limited expression in

Tumor Targeting with Novel 6‑Substituted Pyrrolo [2,3‑d] Pyrimidine Antifolates with Heteroatom Bridge Substitutions via Cellular Uptake by Folate Receptor α and the Proton-Coupled Folate Transporter and Inhibition of de Novo Purine Nucleotide Biosynthesis

open access: yes, 2016
Targeted antifolates with heteroatom replacements of the carbon vicinal to the phenyl ring in 1 by N (4), O (8), or S (9), or with N-substituted formyl (5), acetyl (6), or trifluoroacetyl (7) moieties, were synthesized and tested for selective cellular ...

Targeted Protein Degradation: From Molecular Mechanisms to Emerging Therapeutic Strategies

open access: yesMedComm, Volume 7, Issue 11, November 2026.
Graphical overview of targeted protein degradation: mechanisms, molecular determinants, delivery strategies, and therapeutic development. This graphical abstract summarizes the major degradation pathways involved in targeted protein degradation, including the ubiquitin–proteasome system, endocytosis–lysosome pathway, and autophagy‐dependent degradation.

Combating Campylobacter Resistance in Moroccan Poultry Using Essential Oil–Ciprofloxacin Combination

open access: yesVeterinary Medicine and Science, Volume 12, Issue 6, November 2026.
With an estimated prevalence of 15.52% in Moroccan poultry, Campylobacter species pose a significant food safety concern. We investigated whether essential oils (EOs) could restore the efficacy of ciprofloxacin against multidrug‐resistant (MDR) strains.

A P425R mutation of the proton-coupled folate transporter causing hereditary folate malabsorption produces a highly selective alteration in folate binding

open access: yesAmerican Journal of Physiology-Cell Physiology, 2012
Proton-coupled folate transporter (PCFT) mediates folate intestinal absorption and transport across the choroid plexus, processes defective in subjects with hereditary folate malabsorption (HFM). PCFT is also widely expressed in human solid tumors where it contributes to the transport of pemetrexed and other antifolates.

The role of l-leucovorin uptake and metabolism in the modulation of 5-fluorouracil efficacy and antifolate toxicity

open access: yesFrontiers in Pharmacology
BackgroundL-Leucovorin (l-LV; 5-formyltetrahydrofolate, folinic acid) is a precursor for 5,10-methylenetetrahydrofolate (5,10-CH2-THF), which is important for the potentiation of the antitumor activity of 5-fluorouracil (5FU).

A MUC13 Variant Enhances Intestinal Ferric Iron Absorption in Unexplained Hyperferritinemia

open access: yesLiver International, Volume 46, Issue 11, November 2026.
ABSTRACT Background & Aims Hyperferritinemia frequently remains unexplained despite comprehensive evaluation. We conducted a sequencing‐based exome association study to identify genetic variants associated with plasma ferritin levels in patients with unexplained hyperferritinemia.
