Antibiotics and RNase P

open access: yesAntibiotics, 2016
RNase P is an essential endonuclease in tRNA biogenesis, which generates the mature 5′-termini of tRNAs. Most forms of RNase P are ribonucleoproteins, i.e., they consist of an essential RNA and protein subunits.

An Efficient Synthetic Strategy for the Preparation of Nucleic Acid-Encoded Peptide and Protein Libraries for In Vitro Evolution Protocols

open access: yesMolecules, 2000
We describe an improved synthetic strategy for the preparation of nucleic acid encoded peptide and protein libraries. A solid-phase format was used to prepare and purify a novel type of mRNA-template for in vitro mRNA-protein fusion synthesis.

Adrenalectomy and the Suppression of Memory by Puromycin [PDF]

open access: yesProceedings of the National Academy of Sciences, 1970
It has previously been shown that expression of memory of maze-learning in mice is blocked by puromycin injected intracerebrally one or more days after the training experience. Bilateral adrenalectomy before training has now been found to protect memory against the effects of puromycin. This protection is absent when adrenalectomy follows training.

Targeting of herpes simplex virus 1 thymidine kinase gene sequences into the OCT4 locus of human induced pluripotent stem cells. [PDF]

open access: yesPLoS ONE, 2013
The in vitro differentiation of human induced pluripotent stem cells (hiPSC) to generate specific types of cells is inefficient, and the remaining undifferentiated cells may form teratomas.

Construction of Fosmid-based SARS-CoV-2 replicons for antiviral drug screening and replication analyses in biosafety level 2 facilities

open access: yesVirus Research, 2023
The coronavirus disease 2019 (COVID-19) pandemic, caused by severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2), has necessitated the global development of countermeasures since its outbreak.

Induction of P-glycoprotein overexpression in brain endothelial cells as a model to study blood-brain barrier efflux transport

open access: yesFrontiers in Drug Delivery
The blood-brain barrier (BBB) is comprised of specialized brain endothelial cells (BECs) that contribute to maintaining central nervous system (CNS) homeostasis.

A Microplate-Based Nonradioactive Protein Synthesis Assay: Application to TRAIL Sensitization by Protein Synthesis Inhibitors.

open access: yesPLoS ONE, 2016
Non-radioactive assays based on incorporation of puromycin into newly synthesized proteins and subsequent detection using anti-puromycin antibodies have been previously reported and well-validated.

Puromycin-terminated proteins colocalize with focal adhesion kinase (FAK).

open access: yes, 2018
Confocal microscopy of synovial fibroblasts that were re-adhered for 60 and 120 min, fed with puromycin, double stained using anti-puromycin and FAK antibodies, followed by visualization with Alexa 488 and 594 secondary antibodies, showing colocalization

Effect of different PPT lengths on puromycin sensitivity.

open access: yes, 2022
A. Puromycin sensitivity curves for trypanosomes containing pHD3180 (no boxB) with different PPT lengths. Numbers of viable cells remaining after 48 h (as measured using rezazurin) relative to untreated cells are shown on the y-axis, and the puromycin ...

Puromycin-terminated proteins are detected at the cell membrane in the presence of ADAM15.

open access: yes, 2018
(A) chondrocytes transfected with full-length ADAM15 (+) and the deletion mutant of ADAM15 lacking the cytoplasmic domain (ΔC) were detached and re-adhered for 45 and 60 min, fed with puromycin, crosslinked with PFA and cell surface biotinylated.
