Current Cell/Organoid and Animal Models for Primary Sclerosing Cholangitis
Primary sclerosing cholangitis (PSC) is a chronic cholestatic liver disease with limited therapeutic options and a marked risk of progression to biliary fibrosis, cirrhosis, and malignancy. Progress in PSC research has been hindered by the lack of models that faithfully recapitulate the complex biliary microenvironment and disease heterogeneity ...

Pregnane X Receptor Activation in Liver Macrophages Protects against Endotoxin‐Induced Liver Injury
Endotoxemia‐related acute liver injury has a poor prognosis and high mortality, and macrophage polarization plays a central role in the pathological process.

An oxygen‐responsive living bioreactor was engineered via interspecies protoplast fusion, converting oxygen from an inhibitory stress into a programmable signal for sustained indole‐3‐propionic acid (IPA) production. This genetically encoded bioreactor enables robust aerobic operation and therapeutic efficacy of deoxynivalenol detoxification in vivo ...

The emerging role of the Hippo signaling pathway in interorgan crosstalk
Hippo signaling functions as a central hub of interorgan communication. Systemic cues from the gut, adipose tissue, and skeletal muscle—including hormones, metabolites, and microbial signals—regulate YAP/TAZ activity in a tissue‐ and context‐dependent manner.

Novel functions of PXR in cardiometabolic disease [PDF]
Cardiometabolic disease emerges as a worldwide epidemic and there is urgent need to understand the molecular mechanisms underlying this chronic disease. The chemical environment to which we are exposed has significantly changed in the past few decades and recent research has implicated its contribution to the development of many chronic human diseases.

Elevated circulating pregnane X receptor as a novel diagnostic biomarker in hepatocellular carcinoma [PDF]
Aim: Hepatocellular carcinoma (HCC) poses a significant global health threat. The pregnane X receptor (PXR) is a central regulator of xenobiotic metabolism and plays a key role in mediating cellular resistance to anti-tumor drugs in HCC.

Glucocorticoids modulate drug transporter function in human fetal brain endothelial cells
Abstract figure legend P‐glycoprotein and breast cancer resistance protein are the most prominent drug transporters at the fetal blood–brain barrier. We isolated primary human fetal brain endothelial cells from early and mid‐gestation cerebral microvessels and exposed them to glucocorticoids cortisol and dexamethasone in vitro.

The intestinal functions of PXR and CAR
Pregnane X receptor (PXR) and constitutive androstane receptor (CAR) are so-called xenobiotic nuclear receptors that play pivotal roles in xenobiotic metabolism and detoxification. Both receptors, highly expressed in the liver and intestine, also have endobiotic functions by regulating the homeostasis of endogenous chemicals.

PXR activation impairs hepatic glucose metabolism partly via inhibiting the HNF4α–GLUT2 pathway
Drug-induced hyperglycemia/diabetes is a global issue. Some drugs induce hyperglycemia by activating the pregnane X receptor (PXR), but the mechanism is unclear.
