SINGLE CELL HIGH-THROUGHPUT QRT-PCR PROTOCOL v2 [PDF]

open access: yes, 2020
Single cell high-throughput qRT-PCR protocol combines high sensitivity technique of single cell qPCR with high-throughput qPCR technology that can generate data from 96 samples and 96 genes in a single experiment. It can be adapted for various sample types- cell culture, tissue samples and extracted RNA (10pg) and measured on traditional qPCR and high ...

Relationship Between Neurologic Symptoms and Signs and FMR1 Genotype in Premutation Carriers

open access: yesAnnals of Clinical and Translational Neurology, EarlyView.
ABSTRACT Background and Objectives Fragile X‐associated Tremor/Ataxia Syndrome (FXTAS) is the most severe late‐onset condition caused by a premutation in the FMR1 gene, characterized by expanded CGG triplet repeats of 55–200. Clinical presentations of FXTAS, including gait ataxia, kinetic tremor, cognitive decline, and rare Parkinsonism, are linked to ...

Mechanoregulatory Effects of Cell‐Scale Microwells on Epithelial Cell Phenotype

open access: yesAdvanced Functional Materials, EarlyView.
In small polycaprolactone microwells, A549 epithelial cells span well edges, in contrast to cells growing on flat substrates. Focal adhesion sites (yellow) concentrate at topographic boundaries, while cytoskeletal tension (magenta stress fibers) is transmitted to the nucleus (blue), reducing nuclear sphericity.

Tomato transcriptome and mutant analyses suggest a role for plant stress hormones in the interaction between fruit and Botrytis cinerea. [PDF]

open access: yes, 2013
Fruit-pathogen interactions are a valuable biological system to study the role of plant development in the transition from resistance to susceptibility. In general, unripe fruit are resistant to pathogen infection but become increasingly more susceptible

Microenvironmental Reprogramming by 3D Anisotropic Cardiac Extracellular Matrix Induces Nuclear Remodeling and Epigenetic Maturation of Chemically Induced Cardiomyocytes

open access: yesAdvanced Functional Materials, EarlyView.
A 3D anisotropic hydrogel derived from heart extracellular matrix guides cytoskeletal alignment and nuclear remodeling in reprogrammed cardiomyocyte‐like cells. This study reveals how matrix alignment modulates nuclear envelope dynamics and chromatin state, triggering transcriptional and functional maturation.

Bioactive Conductive Ti3C2Tx‐Ce Hydrogel Facilitates Spinal Cord Injury Repair Through ROS Scavenging and Mitochondrial Regulation

open access: yesAdvanced Functional Materials, EarlyView.
Bioactive conductive Ti3C2Tx‐Ce hydrogels with with scavenging ROS and alleviating neuronal mitochondrial dysfunction are exploited for SCI treatment. This functionality is attributed to interfacial activation modification of Ti3C2Tx with Ce3+ ions to in‐situ form Ce(OH)x‐mediated protective layer, which is beneficial to enabling stable intracellular ...

Multi-Fluorescence Real-Time PCR Assay for Detection of RIF & INH Resistance of M. tuberculosis

open access: yesFrontiers in Microbiology, 2016
Background: Failure to early detect multidrug-resistant tuberculosis (MDR-TB) results in treatment failure and poor clinical outcomes, and highlights the need to rapidly detect resistance to rifampicin (RIF) and isoniazid (INH)Methods: In Multi ...

Self‐Assembled Nanoclay Gel With Spheroid MSC‐Derived Exosome Mimetics to Integrate Demineralized Bone Matrix and Noggin‐Targeting miRNA for Synergistic Osteogenesis

open access: yesAdvanced Functional Materials, EarlyView.
We report a versatile nanoclay gel carrier platform that enhances the osteogenic efficacy of demineralized bone matrix (DBM) by integrating nanoclay‐based self‐assembly with cell‐derived exosome mimetics loaded with noggin‐targeting miR‐200c. This design amplifies BMP signaling, thereby promoting DBM‐mediated osteogenesis and offering a promising ...
