Implementation of Quantitative and Systems Pharmacology in Large Pharma

open access: yesCPT: Pharmacometrics & Systems Pharmacology, 2014
Quantitative and systems pharmacology concepts and tools are the foundation of the model‐informed drug development paradigm at Merck for integrating knowledge, enabling decisions, and enhancing submissions. Rigorous prioritization of modeling and simulation activities has enabled key drug development decisions and led to a high return on investment ...

PDGF regulates guanylate cyclase expression and cGMP signaling in vascular smooth muscle

open access: yesCommunications Biology, 2022
PDGF reduces expression of nitric oxide-sensitive soluble guanylyl cyclase (NO-sGC) through PI3K-P-Rex1-Rac1 signaling in vascular smooth muscle cells.

Quantitative clinical pharmacology and patient-centered healthcare technologies: perspectives 2030

open access: yesРегуляторные исследования и экспертиза лекарственных средств, 2022
One of the most promising trends in clinical pharmacology is pharmacometrics, a combination of pharmacology and statistics that implements quantitative approaches for characterising dose–response relationships and predicting the variability of these ...

Methodologies for Quantitative Systems Pharmacology (QSP) Models: Design and Estimation. [PDF]

open access: yesCPT: pharmacometrics & systems pharmacology, 2017
With the increased interest in the application of quantitative systems pharmacology (QSP) models within medicine research and development, there is an increasing need to formalize model development and verification aspects. In February 2016, a workshop was held at Roche Pharma Research and Early Development to focus discussions on two critical ...

Organ‐specific redox imbalances in spinal muscular atrophy mice are partially rescued by SMN antisense oligonucleotides

open access: yesFEBS Letters, EarlyView.
We identified a systemic, progressive loss of protein S‐glutathionylation—detected by nonreducing western blotting—alongside dysregulation of glutathione‐cycle enzymes in both neuronal and peripheral tissues of Taiwanese SMA mice. These alterations were partially rescued by SMN antisense oligonucleotide therapy, revealing persistent redox imbalance as ...

Transferrin receptor 1‐mediated iron uptake supports thermogenic activation in human cervical‐derived adipocytes

open access: yesFEBS Letters, EarlyView.
In this study, we found that human cervical‐derived adipocytes maintain intracellular iron level by regulating the expression of iron transport‐related proteins during adrenergic stimulation. Melanotransferrin is predicted to interact with transferrin receptor 1 based on in silico analysis.

Modeling and Simulation of Acetaminophen Pharmacokinetics and Hepatic Biomarkers After Overdoses of Extended‐Release and Immediate‐Release Formulations in Healthy Adults Using the Quantitative Systems Toxicology Software Platform DILIsym

open access: yesCPT: Pharmacometrics & Systems Pharmacology
Acetaminophen (APAP) has been formulated as immediate‐, modified‐, and extended‐release tablets (APAP‐IR, ‐MR, and ‐ER, respectively). However, there was concern that APAP‐MR previously available in Europe could form a bezoar after a large overdose ...

In vitro to clinical translation of combinatorial effects of doxorubicin and dexrazoxane in breast cancer: a mechanism-based pharmacokinetic/pharmacodynamic modeling approach

open access: yesFrontiers in Pharmacology, 2023
Dexrazoxane (DEX) is the only drug clinically approved to treat Doxorubicin-induced cardiotoxicity (DIC), however its impact on the anticancer efficacy of DOX is not extensively studied.

Valosin‐containing protein counteracts ATP‐driven dissolution of FUS condensates through its ATPase activity in vitro

open access: yesFEBS Letters, EarlyView.
Biomolecular condensates formed by fused in sarcoma (FUS) are dissolved by high ATP concentrations yet persist in cells. Using a reconstituted system, we demonstrate that valosin‐containing protein (VCP), an AAA+ ATPase, counteracts ATP‐driven dissolution of FUS condensates through its D2 ATPase activity.
