Retinal Progenitor Cells — Timing Is Everything

New England Journal of Medicine, 2007
Permitting progenitor photoreceptor cells from donors to differentiate before transplanting them is key to their successful integration into the retina.

Microscopic characterization of rat retinal progenitor cells

Brain Research, 2007
A progenitor cell line was developed from a postnatal day 2 (P2) rat retina to study the effects of secreted proteins of the retinal pigment epithelium (RPE) on isolated retinal progenitor cells and markers for immature and differentiated retinal cells. Progenitor cells were cloned from a P2 explant grown in secreted proteins of cultured RPE cells.

Growth kinetics and transplantation of human retinal progenitor cells

Experimental Eye Research, 2009
We studied the growth kinetics of human retinal progenitor cells (hRPCs) isolated from donor tissue of different gestational ages (G.A.), determined whether hRPCs can be differentiated into mature photoreceptors and assessed their ability to integrate with degenerating host retina upon transplantation.

Human Retinal Progenitor Cell Transplantation Preserves Vision* [PDF]

open access: yesJournal of Biological Chemistry, 2014
Cell transplantation is a potential therapeutic strategy for retinal degenerative diseases involving the loss of photoreceptors. However, it faces challenges to clinical translation due to safety concerns and a limited supply of cells. Human retinal progenitor cells (hRPCs) from fetal neural retina are expandable in vitro and maintain an ...

Muller glia induce retinal progenitor cells to differentiate into retinal ganglion cells

NeuroReport, 2006
Retinal progenitor cells could differentiate into various retinal cells that made cell-replacement therapy possible. Here, we investigated the role of cellular microenvironment on their regulation and differentiation and found that the percentage of proliferating cells and the percentage of retinal ganglion cells produced from them increased, when ...

E13.5 retinal progenitors induce mouse bone marrow mesenchymal stromal cells to differentiate into retinal progenitor-like cells

Cytotherapy, 2011
Retinal progenitor cells (RPC) are an excellent resource for retinal replacement therapy but usually unavailable. We attempted to induce bone marrow mesenchymal stromal cells (BMSC) into RPC.BMSC and embryonic day 13.5 (E13.5) RPC derived from wild-type or enhanced green fluorescence protein (EGFP) transgenic (Egfp(+/+)) mice were co-cultured in a ...

Influence of a retinal pigment epithelial cell factor(s) on rat retinal progenitor cells

Developmental Brain Research, 1996
Retinal development was studied by explant culture of retinas from embryonic and neonatal rats in response to medium conditioned (CM) by a transformed neonatal rat retinal pigment epithelial (tnrRPE) cell line. Retinal explants from embryonic days 16 and 18 and postnatal day 2 Long-Evans rats were cultured for over 3 weeks on a poly-D,L-ornithine ...

Cell-intrinsic regulators of proliferation in vertebrate retinal progenitors

Seminars in Cell & Developmental Biology, 2004
The proliferative expansion of retinal progenitor cells (RPCs) is a fundamental mechanism of growth during vertebrate retinal development. Over the past couple of years, significant progress has been made in identifying genes expressed in RPCs that are essential for their proliferation, and the molecular mechanisms are beginning to be resolved. In this

Retinal Progenitor Cells, Differentiation, and Barriers to Cell Cycle Reentry

2010
Neurogenesis in the retina occurs via the coordination of proliferation, cell cycle exit and differentiation of retinal progenitor cells. Until recently, it was widely assumed that once a retinal progenitor cell produced a postmitotic neuron, there was no possibility for cell-cycle re-entry. However, recent studies have shown that mature differentiated

Decellularized retinal matrix: Natural platforms for human retinal progenitor cell culture

Acta Biomaterialia, 2016
Tissue decellularization strategies have enabled engineering of scaffolds that preserve native extracellular matrix (ECM) structure and composition. In this study, we developed decellularized retina (decell-retina) thin films. We hypothesized that these films, mimicking the retina niche, would promote human retinal progenitor cell (hRPC) attachment ...
