Rho GTPase Cdc42 is essential for human T-cell development

open access: yesHaematologica, 2010
Background Rho GTPases are involved in the regulation of many cell functions, including some related to the actin cytoskeleton. Different Rho GTPases have been shown to be important for T-cell development in mice.

TRP Channels and Small GTPases Interplay in the Main Hallmarks of Metastatic Cancer

open access: yesFrontiers in Pharmacology, 2020
Transient Receptor Potential (TRP) cations channels, as key regulators of intracellular calcium homeostasis, play a central role in the essential hallmarks of cancer.

Emerging role of ARHGAP29 in melanoma cell phenotype switching

open access: yesMolecular Oncology, EarlyView.
This study gives first insights into the role of ARHGAP29 in malignant melanoma. ARHGAP29 was revealed to be connected to tumor cell plasticity, promoting a mesenchymal‐like, invasive phenotype and driving tumor progression. Further, it modulates cell spreading by influencing RhoA/ROCK signaling and affects SMAD2 activity. Rho GTPase‐activating protein

Deregulation of Rho GTPases in cancer

open access: yesSmall GTPases, 2016
In vitro and in vivo studies and evidence from human tumors have long implicated Rho GTPase signaling in the formation and dissemination of a range of cancers.

In vitro models of cancer‐associated fibroblast heterogeneity uncover subtype‐specific effects of CRISPR perturbations

open access: yesMolecular Oncology, EarlyView.
Development of therapies targeting cancer‐associated fibroblasts (CAFs) necessitates preclinical model systems that faithfully represent CAF–tumor biology. We established an in vitro coculture system of patient‐derived pancreatic CAFs and tumor cell lines and demonstrated its recapitulation of primary CAF–tumor biology with single‐cell transcriptomics ...

Actin dynamics controlled by IqgC, a RasGAP at the crossroads between the IQGAP and fungal GAP1 families

open access: yesFEBS Open Bio, EarlyView.
IqgC is a RasGAP from Dictyostelium discoideum. IqgC binds RasG via its RasGAP domain and deactivates it on macroendocytic cups, thereby suppressing the uptake of fluid and particles. IqgC has a positive effect on cell‐substratum adhesion, and its RGCt domain is required for recruitment to ventral foci.

RHO GTPases in cancer: known facts, open questions, and therapeutic challenges.

open access: yesBiochemical Society Transactions, 2018
RHO GTPases have been traditionally associated with protumorigenic functions. While this paradigm is still valid in many cases, recent data have unexpectedly revealed that RHO proteins can also play tumor suppressor roles. RHO signaling elements can also

Diffusion‐based size determination of solute particles: a method adapted for postsynaptic proteins

open access: yesFEBS Open Bio, EarlyView.
We present a diffusion‐based approach for measuring the size of macromolecules and their complexes, and demonstrate its use on postsynaptic proteins. The method requires fluorescein‐labelled protein samples, a microfluidic device that maintains laminar flow for said samples, a microscope recording the emitted fluorescent signals, and an analytic ...

SmgGDS: An Emerging Master Regulator of Prenylation and Trafficking by Small GTPases in the Ras and Rho Families

open access: yesFrontiers in Molecular Biosciences, 2021
Newly synthesized small GTPases in the Ras and Rho families are prenylated by cytosolic prenyltransferases and then escorted by chaperones to membranes, the nucleus, and other sites where the GTPases participate in a variety of signaling cascades ...

Rho GTPases in Intellectual Disability: From Genetics to Therapeutic Opportunities

open access: yesInternational Journal of Molecular Sciences, 2018
Rho-class small GTPases are implicated in basic cellular processes at nearly all brain developmental steps, from neurogenesis and migration to axon guidance and synaptic plasticity.
