Despite widespread exposure, the mechanisms of manganese neurotoxicity remain poorly understood. Using correlative cryogenic fluorescence and synchrotron X‐ray fluorescence imaging techniques, we show that the Golgi apparatus is the primary accumulation site for manganese in both neurons and astrocytes, and that manganese targets the neuronal ...

Probing the role of the C2F domain of otoferlin
Afferent synapses of cochlear inner hair cells (IHCs) employ a unique molecular machinery. Otoferlin is a key player in this machinery, and its genetic defects cause human auditory synaptopathy.

Synaptic vesicle dynamics in mouse rod bipolar cells. [PDF]
To better understand synaptic signaling at the mammalian rod bipolar cell terminal and pave the way for applying genetic approaches to the study of visual information processing in the mammalian retina, synaptic vesicle dynamics and intraterminal calcium

Neurotransmitter release at ribbon synapses in the retina
The synapses of photoreceptors and bipolar cells in the retina are easily identified ultrastructurally by the presence of synaptic ribbons, electron‐dense bars perpendicular to the plasma membrane at the active zones, extending about 0.5 μm into the cytoplasm.

Molecular dissection of the photoreceptor ribbon synapse [PDF]
The ribbon complex of retinal photoreceptor synapses represents a specialization of the cytomatrix at the active zone (CAZ) present at conventional synapses. In mice deficient for the CAZ protein Bassoon, ribbons are not anchored to the presynaptic membrane but float freely in the cytoplasm.

The ER's continuous tubular network is maintained by ER‐shaping proteins whose mutation or dysregulation contributes to neurodegenerative diseases. Here, we show that ER morphology sets the speed of Ca2+ store replenishment between firing events. Disrupting ER continuity slows intra‐ER Ca2+ redistribution from extracellular refill (SOCE) sites, driving

An Introduction to Predictive Processing Models of Perception and Decision‐Making
Abstract The predictive processing framework includes a broad set of ideas, which might be articulated and developed in a variety of ways, concerning how the brain may leverage predictive models when implementing perception, cognition, decision‐making, and motor control.

New England Overview: A Guide to Large-Scale Energy Infrastructure Issues in 2015 [PDF]
The report outlines how regional electricity and natural gas infrastructure decisions are made. It examines the current proposals to expand electricity transmission lines and natural gas pipelines into New England, as solutions to electricity and gas ...

Photoreceptor Outer Segment-like Structures in Long-Term 3D Retinas from Human Pluripotent Stem Cells. [PDF]
The retinal degenerative diseases, which together constitute a leading cause of hereditary blindness worldwide, are largely untreatable. Development of reliable methods to culture complex retinal tissues from human pluripotent stem cells (hPSCs) could ...

Studying Macromolecular Composition in Cell–Cell Interfaces Using 3D Membrane Reconstitution Systems
A comprehensive understanding of the interactions between proteins, lipids and glycocalyx components at the immune synapse is still lacking. Here, an artificial and a semi‐artificial model contact system were established to reconstitute the cell‐cell contact in 3D. The model systems enable the examination of macromolecule enrichment or depletion at the
