Positional scanning and computational modeling reveal determinants of legumain transpeptidase activity

open access: yesProtein Science, Volume 35, Issue 5, May 2026.
Abstract Legumain is a lysosomal cysteine protease that plays a central role in antigen processing for MHC class II presentation. Beyond its canonical proteolytic activity, legumain can also function as a peptide ligase or transpeptidase. This non‐canonical activity becomes particularly relevant under pathophysiological conditions such as cancer and ...

Function of the Signal Peptide and N- and C-terminal Propeptides in the Leucine Aminopeptidase from \u3cem\u3eAeromonas proteolytica\u3c/em\u3e [PDF]

open access: yes, 2004
The leucine aminopeptidase from Aeromonas proteolytica (also known as Vibrio proteolyticus) (AAP) is a metalloenzyme with broad substrate specificity. The open reading frame (ORF) for AAP encodes a 54 kDa enzyme, however, the extracellular enzyme has a ...

Modeling Synaptic Maturation From Growth Cone to Synapse in Human Organoids

open access: yesJournal of Neurochemistry, Volume 170, Issue 5, May 2026.
Synapses are implicated in several neurological disorders and psychiatric diseases. The emergence and wide use of neural organoids provide a new opportunity to study human synapses in healthy and disease settings. Therefore, we developed a simple method for the enrichment of synaptosomes and growth cone particles from forebrain organoids. The method is

Processing and release of endogenous immunogenic peptide signals

open access: yesThe Plant Journal, Volume 126, Issue 3, May 2026.
SUMMARY In both animals and plants, small secreted peptides known as cytokines and phytocytokines mediate local and systemic communication during immune and stress responses. These signaling molecules are typically synthesized as inactive precursors that require proteolytic processing to become active.

The functions of DNA methyltransferases during the feeding and development of Haemaphysalis longicornis are potentially associated with lysosome pathways

open access: yesBMC Genomics
Background DNA methylation is an epigenetic modification that plays an important role in animal and plant development. Among the diverse types of DNA methylation modifications, methylation of cytosines catalyzed by DNA cytosine methyltransferases (DNMTs)

Safety evaluation of the food enzyme containing aspergillopepsin I and carboxypeptidase C activities from the non‐genetically modified Aspergillus sp. strain ACP 112–311

open access: yesEFSA Journal, Volume 24, Issue 5, May 2026.
Abstract The food enzyme with aspergillopepsin I (EC 3.4.23.18) and carboxypeptidase C (EC 3.4.16.5) activities is produced with the non‐genetically modified Aspergillus sp. strain ACP 112–311 by Shin Nihon Chemical Co., Ltd. The food enzyme was considered free from viable cells of the production organism.
