Platelet aggregation and sphingomyelinase D activity of a purified toxin from the venom of loxosceles reclusa

Biochimica et Biophysica Acta (BBA) - General Subjects, 1981
A facile and quantitative assay for measuring the activity of sphingomyelinase D in recluse spider venom has been developed using L-alpha-[palmitoyl-1-14C]lysophosphatidylcholine as substrate. This assay avoids the problem of substrate insolubility that occurs when sphingomyelin and other insoluble lipids are used as substrates.

North and South American Loxosceles spiders: Development of a polyvalent antivenom with recombinant sphingomyelinases D as antigens

Toxicon, 2006
We report the cloning of sphingomyelinase D (SMD) cDNA from Loxosceles reclusa, Loxosceles boneti and Loxosceles laeta into bacterial expression systems, as well as optimization of expression conditions so as to obtain soluble and active recombinant enzymes. The recombinant mature SMDs, tagged with a histidine tail at the N- or C-termini, were compared

Comparative analyses of venoms from American and African Sicarius spiders that differ in sphingomyelinase D activity

Toxicon, 2010
Spider venoms are cocktails of toxic proteins and peptides, whose composition varies at many levels. Understanding patterns of variation in chemistry and bioactivity is fundamental for understanding factors influencing variation. The venom toxin sphingomyelinase D (SMase D) in sicariid spider venom (Loxosceles and Sicarius) causes dermonecrotic lesions

Role of the complement system in kidney cell death induced by Loxosceles venom Sphingomyelinases D

Archives of Toxicology
Envenomation by Loxosceles spiders can result in local and systemic pathologies. Systemic loxoscelism, which can lead to death, is characterized by intravascular hemolysis, platelet aggregation, and acute kidney injury. Sphingomyelinase D (SMase D) in Loxosceles spider venom is responsible for both local and systemic pathologies, and has been shown to ...

Lysosomal Regulation of Gasdermin D Pore Formation in Podocytes Determined by Acid Sphingomyelinase

Physiology
Recent studies have shown that gasdermin D (GSDMD) pore may be involved in inflammasome product release and pyroptosis in podocytes. Nevertheless, the molecular mechanism by which GSDMD pore formation is regulated in these cells remains poorly understood.
