Specific inhibition of Src-Tyrosine kinase reduces STAT6 activation.

open access: yes, 2012
(A) Tip, Lck and STAT6 transfected 293T cells were treated with PP2 or the non-active control substance PP3 (10 µM in DMSO each). STAT6, pSTAT6, Tip and GAPDH were detected by fluorescent immunoblotting on the same membrane.

Schisandrin C Targets PPARγ to Induce Ferroptosis in Colorectal Cancer Cells and Promote M1 Macrophage Polarization

open access: yesPhytotherapy Research, EarlyView.
Schisandrin C: A Dual Anti‐Colorectal Cancer Strategy Targeting PPARγ. ABSTRACT Colorectal cancer (CRC) remains a leading cause of cancer mortality, necessitating novel therapeutic strategies. Schisandrin C (SinC), a bioactive lignan from Schisandra chinensis, exhibits diverse pharmacological activities, but its anti‐CRC mechanisms are poorly ...

Autosomal Dominant Hyper‐IgE Syndrome Patients Retain IL10‐Producing preTh17‐Cells That Are Activated by Opportunistic Pathogens and Support IgE Production

open access: yesAllergy, EarlyView.
IL‐10 producing CCR6+Th‐cells are central memory T‐cells that express ROR‐γt and differentiate to Th17‐cells via an autocrine loop of STAT3‐activating cytokines (preTh17). STAT3‐deficient AD‐HIES patients lack Th17‐ and Tfh17‐cells but retain preTh17‐ and Th1/17‐cells.

Lupus Panniculitis Immunoprofiling Reveals a Cytotoxic, Type 1‐Skewed T‐Cell Response With Signs of Metabolic Alteration

open access: yesAllergy, EarlyView.
Imaging mass cytometry shows lupus panniculitis lesions are dominated by cytotoxic and skin‐homing T cells with abundant macrophages and B‐cell aggregates. Spatial neighborhood analysis reveals organized leukocyte architecture, including B‐cell proximity to naïve T cells and M1 macrophages to cytotoxic T cells.

Modulation of M2 macrophage polarization by the crosstalk between Stat6 and Trim24

open access: yesNature Communications, 2019
Stat6 promotes M2 macrophage polarization. Here the authors characterize Trim24-CBP-Stat6 circuit regulating M2 macrophage polarization via Stat6 acetylation, and show it contributes to pro-tumorigenic macrophage activity in mice.

Stat6 dependent goblet cell hyperplasia during intestinal nematode infection

open access: yes, 2015
To identify the role of signal transducer and activator of transcription factor 6 (Stat6) in the development of intestinal goblet cell hyperplasia during nematode infection, we compared the number of goblet cells in Stat6 deficient (Stat6 -/-) mice with ...

Understanding the Type 2 Immunity: An Evolving View Beyond Allergic Diseases

open access: yesAllergy, EarlyView.
ABSTRACT Type 2 (T2) immunity is classically associated with defense against helminths and environmental threats, from a physiological perspective, and with allergies and eosinophilic inflammatory diseases, in a pathological sense. However, growing evidence reveals that T2 pathways also support other essential homeostatic functions, including tissue ...

Regulation des STAT6/ Koaktivator Komplexes in der Zytokin induzierten Transkription [PDF]

open access: yes, 2010

PLAG inhibits IL-4 induced STAT6 phosphorylation.

open access: yes, 2016
(A) Various concentration of a STAT6 inhibitor (STAT6i, AS1517499) were treated onto A549 cells which were activated by IL-4 (10 ng/mL), and showed inhibitory effect on the secretion of CCL26 from the epithelial cells in a concentration-dependent manner.

T Cell Microenvironmental Impacts and Therapeutic Opportunities: Application to Human Testicular Germ Cell Tumors

open access: yesAndrology, EarlyView.
ABSTRACT Testicular germ cell tumors (TGCT) are the leading malignancy in adolescent and young adult males, yet the immunological and cellular mechanisms governing their tumor microenvironment (TME) remain poorly understood. Here, we present a comprehensive review of TGCT pathobiology with a focus on the immune landscape, particularly the role of tumor‐
