SuFEx‐Enabled Reprogramming of Flavonoids for Selective α‐Glucosidase Covalent Inhibition

open access: yesAdvanced Science, EarlyView.
Selective inhibition of intestinal α‐glucosidase remains limited by poor enzyme specificity and off‐target metabolic effects. Here, SuFEx click chemistry is used to reprogram natural flavonoids into covalent inhibitors with enhanced α‐glucosidase selectivity and controlled reactivity. This strategy enables effective regulation of carbohydrate digestion

Heat Shock Protein 90: From Molecular Chaperone Function to Therapeutic Targeting in Malignancies

open access: yesAdvanced Science, EarlyView.
In this review, an integrated conceptual framework linking HSP90's molecular chaperone functions to its pathological roles in cancer is proposed. HSP90 serves as a central node that integrates oncogenic signaling, buffers proteotoxic stress, maintains cancer stem cell plasticity, and shapes tumor‐immune interactions, all of which converge to drive ...

A Kinetic Basis For T Cell Receptor Repertoire Selection during an Immune Response

open access: yes, 1999
The basis for T cell antigen receptor (TCR) repertoire selection upon repeated antigenic challenge is unclear. We evaluated the avidity and dissociation kinetics of peptide/major histocompatibility complex (MHC) tetramer binding to antigen-specific T ...

Lessons From Drug Discovery for Cryoprotective Agent Design: An AI‐Oriented Perspective

open access: yesAdvanced Science, EarlyView.
Cryoprotectant design is reframed through the lens of drug discovery as a multiparameter optimization problem. This perspective highlights how AI and systematic design strategies could enable safer, more effective cryoprotectants, while identifying key limitations that currently constrain predictive progress in cryobiology. ABSTRACT Cryopreservation is

Indian hedgehog (Ihh) both promotes and restricts thymocyte differentiation

open access: yes, 2009
We show that Indian Hedgehog (Ihh) regulates T-cell development and homeostasis in both fetal and adult thymus, controlling thymocyte number. Fetal Ihh-/- thymi had reduced differentiation to double-positive (DP) cell and reduced cell numbers compared ...

CCL3 and IL‐7 Synergistically Enhance CAR‐T Efficacy in Solid Tumors

open access: yesAdvanced Science, EarlyView.
Synergistic delivery of CCL3 and IL‐7 by CAR‐T cells overcomes the immunosuppressive tumor microenvironment by enhancing cell infiltration, survival, and local immune reprogramming. This also establishes positive feedback that amplifies anti‐tumor activity and memory responses in both CAR‐T and endogenous T cells, presenting a robust therapeutic ...

T-cell receptor excision circles and repertoire diversity in children with profound T-cell immunodeficiency

open access: yes, 2013
Background/Purpose(s)Approximately 40% of patients with profound T-cell immunodeficiency have no identified molecular basis. Early assessment of T-cell impairment is vital for medical intervention, if hematopoietic stem cell transplantation is needed ...

A Conserved DT2‐bZIP66‐NF‐YC4 Regulatory Module Confers Drought Tolerance in Rice and Arabidopsis

open access: yesAdvanced Science, EarlyView.
This study identifies a conserved tripartiteDT2–bZIP66–NF‐YC4 transcriptional module that enhances rice drought tolerance by activating stress‐responsive genes. This regulatory complex is functionally conserved across cereals and Arabidopsis, providing promising targets for engineering drought‐resilient crops. ABSTRACT Drought stress severely restricts

ZNF33B Promotes Japanese Encephalitis Virus Infection by Regulating the Stability of M6A‐Modified Trim25 to Control the Autophagy Process

open access: yesAdvanced Science, EarlyView.
Upon JEV infection, ZNF33B recruits METTL14 to stabilize the METTL3‐METTL14 m6A methyltransferase complex, leading to increased m6A modification of host transcripts, including Trim25 mRNA. ZNF33B selectively binds m6A‐modified sites on Trim25 mRNA and accelerates its decay, resulting in reduced TRIM25 protein abundance.

Mathematical modelling of T cell homeostasis

open access: yes, 2010
T cell homeostasis describes the process through which the immune system regulates cell survival, proliferation, differentiation and death to maintain T cell numbers and diversity in a range of different conditions. The aim of this thesis is to better
