Objective Somatic DNMT3A mutations are the most common drivers of clonal hematopoiesis in patients with rheumatoid arthritis (RA) and have been associated with seropositive disease and increased inflammatory markers. These mutations are predominantly hypomorphic or dominant‐negative, reducing DNMT3A function.

T-cell Receptor (TCR) targeting with Multivalent T-cell Engagers
Abstract T-cell engagers (TCEs) for cancer immunotherapy have traditionally relied on high affinity single chain fragment variable (scFv) domains to target CD3, specifically the ε chain, for the activation of T-cells.

Objective Axial spondyloarthritis (axSpA) and psoriatic arthritis (PsA) are overlapping yet distinct conditions within the spondyloarthritis (SpA) spectrum. As divergent immunophenotypes may influence disease course and therapeutic response, we compared immune cell subsets, cytokine profiles, and inflammatory mediators.

Role and specificity of regulatory T cells during retroviral infection [PDF]
During chronic viral infection of the hematopoietic system, both virus-mediated and immune-mediated effects may cause bone marrow dysfunction leading to cytopenias. To study the pathological consequences of the T helper response to unresolving chronic

Efficacy, safety and cost‐effectiveness of CAR‐T therapy
CAR T‐cells demonstrate high efficacy in blood cancers, including ALL, MM and DLBCL. Innovations target solid tumours despite challenges such as antigen escape. Combination therapies enhance the delivery and infiltration of CAR T cells. Toxicity, cost and resistance remain major barriers to clinical use.

The zinc finger transcription factor Early Growth Response 2 (Egr-2) is an intrinsic regulator of T cell tolerance and homeostasis [PDF]
PhDTolerance of T cells to self-antigen is crucial to prevent the development of autoimmune disease. How self-tolerance is controlled at the transcriptional level is, however, unknown. We discovered that the transcription factor Early Growth Response 2

Targeting HPK1 is a promising strategy to restore T‐cell anti‐tumor immunity. We report the design and synthesis of novel 2,4‐diaminopyrimidine derivatives, focusing on systematic structure–activity relationship (SAR) analysis These findings identify key structural features for potent HPK1 inhibition, facilitating the discovery of next‐generation ...

Mathematical modelling of T cell homeostasis [PDF]
T cell homeostasis describes the process through which the immune system regulates cell survival, proliferation, differentiation and death to maintain T cell numbers and diversity in a range of different conditions. The aim of this thesis is to better
