Genotoxicity of the phosphoramidate agent tabun (GA)

Five mutagenicity tests were performed on Agent GA (Tabun, phosphoramidocyanidic acid, dimethyl-, ethyl ester) as part of a program to demilitarize chemical warfare agents. GA was mutagenic in Salmonella spp. assays with S-9 and it was a direct-acting mutagen to mouse lymphoma cells.

Toxogonin in Sarin, Soman and Tabun poisoning

The oximes P2S, TMB-4 and Toxogonin have been tested as reactivators of Sarin and Tabun inhibited acetylcholinesterase. The protective effect of the oximes has been studied on atropinized mice in experimental Sarin, Soman and Tabun poisoning. Toxogonin is a better reactivator than P2S and comparable to TMB-4. In the presence of 10 mg/kg atropine (i.p.)

INHIBITION OF THE ESTERATIC ACTIVITY OF THROMBIN BY SARIN AND TABUN

Isopropyl methylphosphonofluoridate (sarin) and ethyl N,N-dimethylphosphoramidocyanidate (tabun) have been found to inhibit the esteratic activity of thrombin towards the substrate p-toluenesulphonyl-L-arginine methyl ester. The rate of inhibition is markedly dependent on pH and is greatest in alkaline solutions.

Synthesis of a New Reactivator (I) of Tabun‐Inhibited Acetylcholinesterase.

Synthesis of a new asymmetric bisquaternary reactivator of tabun-inhibited acetylcholinesterase-1-(4-hydroxyiminomethylpyridinium)-4-(4-carbamoylpyridinium) butane dibromide is described. Reactivation potency of this oxime is compared to the currently used reactivators-pralidoxime, obidoxime and H-oxime HI-6.

The enzymes which hydrolyze isopropyl methylphosphonofluoridate (sarin) and ethyl N, N-dimethylphosphoramidocyanidate (tabun) have been studied. Michaelis–Menten constants and activation energies have been estimated and enzyme stability has been studied. The distribution of the enzymes in mammalian tissues has been examined.
