Studies on the biosynthesis of taxol: the taxane carbon skeleton is not of mevalonoid origin. [PDF]

open access: bronzeProceedings of the National Academy of Sciences, 1996
A cell culture of Taxus chinensis was established to produce the diterpene 2alpha,5alpha,10beta,14beta-tetra-acetoxy4 ++ +(20),11-taxadiene (taxuyunnanine C) in 2.6% (dry weight) yield. The incorporation of [U-13C6]glucose, [1-13C]glucose, and [1,2-13C2]acetate into this diterpene was analyzed by NMR spectroscopy.

Fungal endophytes of Taxus species and regulatory effect of two strains on taxol synthesis [PDF]

open access: yesBMC Microbiology
Background Taxol, derived from Taxus trees, is a valuable natural resource for the development of anticancer drugs. Endophytic fungi from Taxus trees are a promising alternative source of Taxol.

Screening of taxol biosynthesis-related genes in taxol produced from Nodulisporium sylviforme HDF-68 by mRNA differential display [PDF]

open access: bronzeAnnals of Microbiology, 2014
Differential expressed genes in taxol producing Nodulisporium sylviforme HDF-68 at different periods of fermentation were analyzed by mRNA differential display. A total of 38 fragments were found expressed differentially during taxol synthesis phase. Using reverse Northern blot analysis, the hybridization signals of 16 fragments were consistent with ...

Amesia nigricolor, a novel endophyte of Encephalartos bubalinus, exhibiting a robust taxol biosynthetic stability: chemical characterization and biological activities [PDF]

open access: yesMicrobial Cell Factories
Diminishing the productivity of Taxol by the potential fungi with storage is the key hurdle that impedes their applications to be an industrial platform for Taxol production.

Biosynthesis of the Red Algal Diterpene Peyssonnosol in Bacteria. [PDF]

open access: yesAngew Chem Int Ed Engl
Two terpene synthases derived from Anaerolineae bacteria catalyse the biosynthesis of peyssonnosene‐type diterpenes from red algae. Site‐directed mutagenesis enabled a partial functional switch, resulting in the formation of additional byproducts. The intricate cyclisation mechanisms were investigated through isotopic labelling experiments and density ...

Studies on Taxol biosynthesis: preparation of taxadiene-diol and triol derivatives by deoxygenation of taxusin [PDF]

open access: greenTetrahedron, 2008
The putative taxol biosynthesis metabolites, taxa-4(20),11(12)-diene-5α, 13α -diol (7), taxa-4(20),11(12)-diene-5α, 9α, 13α-triol (9), and taxa-4(20),11(12)-diene-5α, 10β, 13α-triol (10), have been prepared by Barton deoxygenation of the C-9 and C10-hydroxyl groups of protected derivatives of taxusin, a major taxoid metabolite isolated from Yew heart ...

Transcriptome and Metabolome Analyses of Taxus chinensis var. mairei Tissues Provide New Insights into the Regulation of Paclitaxel Biosynthesis [PDF]

open access: yesPlants
Taxus is the natural source of the anticancer drug paclitaxel. Although significant progress has been made in elucidating the biosynthetic pathway of paclitaxel, its tissue-specific accumulation and associated regulatory networks in Taxus remains unclear.

Taxol biosynthesis: Identification and characterization of two acetyl CoA:taxoid-O-acetyl transferases that divert pathway flux away from Taxol production [PDF]

open access: greenArchives of Biochemistry and Biophysics, 2009
Two cDNAs encoding taxoid-O-acetyl transferases (TAX 9 and TAX 14) were obtained from a previously isolated family of Taxus acyl/aroyl transferase cDNA clones. The recombinant enzymes catalyze the acetylation of taxadien-5alpha,13alpha-diacetoxy-9alpha,10beta-diol to generate taxadien-5alpha,10beta,13alpha-tri-acetoxy-9alpha-ol and taxadien-5alpha ...
