The germline/somatic DNA damage repair gene mutations modulate the therapeutic response in Chinese patients with advanced pancreatic ductal adenocarcinoma

open access: yesJournal of Translational Medicine, 2021
Background Pancreatic ductal adenocarcinoma (PDAC) is a fatal disease with molecular heterogeneity, inducing differences in biological behavior, and therapeutic strategy.

Pre‐Encoded IFN‐I Sensitivity Exacerbates Memory T Cell Senescence in Solid Tumors

open access: yesAdvanced Science, EarlyView.
Type I interferon (IFN‐I) signaling promotes p21‐dependent cell cycle arrest in senescent tumor‐specific memory T cells, resulting in poor proliferative responses and solid tumor regression during cancer vaccination. Conversely, IFNα/β receptor blockade reinvigorates T cell proliferation to regress solid tumors and is more effective with increasing ...

Homoisoflavanone Delays Colorectal Cancer Progression via DNA Damage‐Induced Mitochondrial Apoptosis and Parthanatos‐Like Cell Death

open access: yesAdvanced Science, EarlyView.
Homoisoflavanone (HIF), a bioactive compound isolated from Polygonatum kingianum, selectively suppresses colorectal cancer progression by inducing DNA damage‐mediated mitochondrial apoptosis and parthanatos‐like cell death. HIF triggers mitochondrial dysfunction, including depolarized membrane potential, elevated ROS, and ATP depletion, while impairing

Targeting WEE1 in ARID1A/TP53 Concurrent Mutant Colorectal Cancer by Exploiting R‐Loop Accumulation and DNA Repair Deficiencies

open access: yesAdvanced Science, EarlyView.
ARID1A, a SWI/SNF complex component, is frequently mutated in colorectal cancer (CRC). CRC with ARID1A/TP53 concurrent mutations shows marked sensitivity to WEE1 inhibition. ARID1A loss induces R‐loop‐mediated replication stress, impairs ATF3 transcription, and amplifies WEE1i‐induced DNA damage, suggesting a promising therapeutic vulnerability ...

Disarmament, demobilisation and reintegration (DDR) as a participatory process: involving communities and beneficiaries in post-conflict disarmament programmes [PDF]

open access: yes, 2008
Disarmament, Demobilisation and Reintegration (DDR) programmes are a structured approach to returning soldiers and militia members to civilian life. One of the aims is to support implementation of the peace process, by addressing their interests and ...

Altered modulation of lamin A/C-HDAC2 interaction and p21 expression during oxidative stress response in HGPS [PDF]

open access: yes, 2018
Defects in stress response are main determinants of cellular senescence and organism aging. In fibroblasts from patients affected by Hutchinson-Gilford progeria, a severe LMNA-linked syndrome associated with bone resorption, cardiovascular disorders, and

Engineering Osteoimmune Responses with Functionalized Orthopedic Implants for Post‐Operative Osteosarcoma Treatment

open access: yesAdvanced Science, EarlyView.
Osteosarcoma is the most common primary bone tumor with limited treatment options and a terrible prognosis. This review provides a comprehensive summary of the recent development of osteoimmunomodulatory implants for post‐operative osteosarcoma treatment, of which the potential utility in evoking durable anti‐osteosarcoma immunity and accelerating bone

Trust and glycemic control in black patients with diabetic retinopathy: A pilot study [PDF]

open access: yes, 2019
Diabetic retinopathy (DR) is more prevalent in blacks than whites because, compared to whites, blacks on average have worse glycemic control. Both of these racial disparities reflect differences in sociocultural determinants of health, including ...

Low YTHDC1 Expression Upregulates FSCN1 to Promote Nuclear F‐Actin Formation and Facilitate Double‐strand DNA Breaks Repair in TMZ‐Resistant Glioblastoma

open access: yesAdvanced Science, EarlyView.
This study revealed that low expression of YTHDC1 in TMZ‐resistant GBM cells leads to increased FSCN1 expression, which is suppressed by m6A modification. FSCN1 activates the CDC42/N‐WASP/Arp2/3 axis in the cell nucleus by recruiting the guanine nucleotide exchange factor (GEF) family member FGD1, thereby promoting F‐actin polymerization in the nucleus.

DNA damage response alterations in clear cell renal cell carcinoma: clinical, molecular, and prognostic implications

open access: yesEuropean Journal of Medical Research
Background DNA damage repair (DDR) pathways modulate cancer risk, progression, and therapeutic responses. Nonetheless, the characteristics and significance of DDR alterations in clear cell renal cell carcinoma (ccRCC) remain undefined.
