Chronobiology of Cancer: How Aging Fuels Oncogenesis at the Molecular Level

open access: yesAging and Cancer, EarlyView.
This graphical abstract illustrates the key biological pathways linking aging with cancer development and progression. In the upper left, cumulative exposure to ultraviolet radiation, toxins, and reactive oxygen species (ROS) causes DNA damage and genomic instability, whereas age‐related decline in repair mechanisms, such as ATM/ATR, BER, and NER ...

Generation of Functional Patient-Specific Thymus Organoids From Human Pluripotent Stem Cells (hPSCs) Using Air–Liquid Interface Culture

open access: yesBio-Protocol
The thymus is critical for the establishment of a functional and self-tolerant adaptive immune system, but it involutes with age, resulting in reduced naive T-cell output.

Mesenchymal cell replacement corrects thymic hypoplasia in murine models of 22q11.2 deletion syndrome

open access: yesThe Journal of Clinical Investigation, 2023
22q11.2 deletion syndrome (22q11.2DS) is the most common human chromosomal microdeletion, causing developmentally linked congenital malformations, thymic hypoplasia, hypoparathyroidism, and/or cardiac defects.

A Novel IDO1/NE Dual Inhibitor, IMM‐H018 Prevents the Primary and Secondary Sepsis and Ameliorates the Kidney Injury Through Inhibiting the Cytokine Storm and Microthrombosis, and Reversing Immunosuppression

open access: yesAdvanced Science, EarlyView.
IMM‐H018, a dual IDO1/NE inhibitor, demonstrates potent therapeutic efficacy in sepsis by simultaneously targeting excessive inflammation and immune dysfunction. It prevents both primary and secondary sepsis through anti‐inflammatory, immune‐restoring, and renoprotective mechanisms, reducing organ damage, improving immune homeostasis, preserving kidney

Thymic Stromal Lymphopoietin–Driven B-Cell Hyperactivation Impairs Intestinal Epithelial Homeostasis Predisposing to Enterocolitis in Hirschsprung DiseaseSummaryWhat You Need to Know

open access: yesCellular and Molecular Gastroenterology and Hepatology
Background & Aims: Hirschsprung-associated enterocolitis, a lethal complication of Hirschsprung’s disease—a prevalent congenital disorder with high morbidity—has a poorly understood pathogenesis.

Genetic Ablation and Multi‐Omics Profiling Reveal CEP55 as a Key Driver of Tumorigenesis in Diverse Cancer Models

open access: yesAdvanced Science, EarlyView.
Genetic ablation of Cep55 in Pten‐deficient mouse models delays tumorigenesis. Integrated multi‐omics analyses (proteomics, phosphoproteomics, and spatial transcriptomics) reveal that CEP55 regulates oncogenic signaling (RAS/ERK, PI3K/AKT), integrin/FAK‐mediated adhesion, extracellular matrix (ECM) remodeling, and endocytosis.

Thymic homing of activated CD4+ T cells induces degeneration of the thymic epithelium through excessive RANK signaling

open access: yesScientific Reports, 2017
Activated T cells have been shown to be able to recirculate into the thymus from the periphery. The present study was aimed to elucidate the functional consequences of thymic homing of activated T cells upon developing thymocytes and thymic epithelial ...

Thymospheres Are Formed by Mesenchymal Cells with the Potential to Generate Adipocytes, but Not Epithelial Cells

open access: yesCell Reports, 2017
Summary: Evidence suggests that a stem-cell-driven differentiation hierarchy maintains the dynamic thymic epithelial cell (TEC) network that governs T lymphocyte development.

Autophagy in the thymic epithelium is dispensable for the development of self-tolerance in a novel mouse model. [PDF]

open access: yesPLoS ONE, 2012
The thymic epithelium plays critical roles in the positive and negative selection of T cells. Recently, it was proposed that autophagy in thymic epithelial cells is essential for the induction of T cell tolerance to self antigens and thus for the ...

Interleukin 7 from maternal milk crosses the intestinal barrier and modulates T-cell development in offspring [PDF]

open access: yes, 2011
Background Breastfeeding protects against illnesses and death in hazardous environments, an effect partly mediated by improved immune function. One hypothesis suggests that factors within milk supplement the inadequate immune response of the offspring,
