Three-Dimensional Heterocycles: New Uracil-Based Structures Obtained by Nucleophilic Substitution at the sp2 Carbon of Bromoisoxazoline

open access: yesMolecules, 2014
The regioisomeric cycloadducts of bromonitrile oxide and N-benzoyl-2,3-oxaza-norborn-5-ene were easily prepared and elaborated into a novel class of uracil-based scaffolds. The key-synthetic step is the nucleophilic substitution at the sp2 carbon atom of

dUTPase is essential in zebrafish development and possesses several single‐nucleotide variants with pronounced structural and functional consequences

open access: yesFEBS Open Bio, EarlyView.
dUTPases are involved in balancing the appropriate nucleotide pools. We showed that dUTPase is essential for normal development in zebrafish. The different zebrafish genomes contain several single‐nucleotide variations (SNPs) of the dut gene. One of the dUTPase variants displayed drastically lower protein stability and catalytic efficiency as compared ...

Harnessing Methyltransferase‐Guided Targeting for Sequence‐Specific Proximity Labeling of DNA

open access: yesAngewandte Chemie, EarlyView.
A novel methyltransferase‐mediated approach leveraging rationally designed S‐adenosyl‐L‐methionine (SAM) analogues was developed to achieve sequence‐specific DNA labeling but in proximity to the natural transfer site, addressing key limitations of previously established methods, including ligand instability and methylation‐sensitive labeling.

Expression and the Peculiar Enzymatic Behavior of the Trypanosoma cruzi NTH1 DNA Glycosylase. [PDF]

open access: yesPLoS ONE, 2016
Trypanosoma cruzi, the etiological agent of Chagas' disease, presents three cellular forms (trypomastigotes, epimastigotes and amastigotes), all of which are submitted to oxidative species in its hosts. However, T.

Combinatorial Synthesis of Next Generation Water‐Soluble Quaternized N‐Halamine Oligomers with Long‐Lasting Antiviral Properties

open access: yesAdvanced Functional Materials, EarlyView.
A combinatorial library of dual‐functional antiviral oligomers incorporating N‐halamine and quaternary ammonium functionalities is developed for long‐lasting antiviral activity. The lead materials exhibit rapid and durable antiviral activity against SARS‐CoV‐2 variants and influenza H1N1, with 4 to 5 log reduction in viral copies at 5 mg mL−1 ...

Biosynthesis of Kaitocephalin: A Neuroprotective Natural Product Featuring a Peptide‐Like yet Nonpeptidic Scaffold

open access: yesAngewandte Chemie, EarlyView.
Kaitocephalin (KCP) is a neuroprotective fungal metabolite with a unique scaffold of amino acids linked via C–C bonds. Genome‐transcriptome analyses identified its biosynthetic gene cluster (kpb cluster) in Eupenicillium shearii. LC‐MS/MS profiling identified four new KCP‐related compounds.

Molecularly Engineered Highly Stable Memristors with Ultra‐Low Operational Voltage: Integrating Synthetic DNA with Quasi‐2D Perovskites

open access: yesAdvanced Functional Materials, EarlyView.
Molecularly engineered memristors integrating Ag nanoparticle–embedded synthetic DNA with quasi‐2D halide perovskites enable ultra‐low‐operational voltage, forming‐free resistive switching, and record‐low power density. This synergistic integration of customized DNA and 2D OHP in bio‐hybrid architecture enhances charge transport, reduces variability ...

Dissipatively Fueled Unidirectionally Communicating DNA Circuits That Control Biocatalysis

open access: yesAngewandte Chemie, EarlyView.
Unidirectionally communicating, out‐of‐equilibrium DNA circuits are used to control enzyme activity. Transient DNAzyme activation generates fuel for temporal trypsin activation. Cyclic fuel‐driven hybridization and exonuclease‐mediated reset enable precise control over lifetimes, which is analyzed using combined experimental and computational ...

Photocrosslinking Probes Proximity of Thymine Modifiers Tethering Excitonically Coupled Dye Aggregates to DNA Holliday Junction

open access: yesMolecules, 2022
A DNA Holliday junction (HJ) has been used as a versatile scaffold to create a variety of covalently templated molecular dye aggregates exhibiting strong excitonic coupling. In these dye-DNA constructs, one way to attach dyes to DNA is to tether them via

Decoupling Dynamics and Crosslink Stability in Supramolecular Hydrogels Using Associative Exchange

open access: yesAdvanced Materials, EarlyView.
How does reorganization impact the stability of supramolecular hydrogels? In contrast to dissociative crosslink exchange, implementing associative exchange in macroscale DNA hydrogels enables decoupling of reorganization dynamics from thermal and mechanical stability.
