M2-polarized macrophages and interleukin (IL)-6 significantly alter the tumor microenvironment and promote the malignant behaviors of tumor cells. This study aimed to establish M2-type macrophages from THP-1 cells, which are human leukemia monocytes, and

Role of Antimicrobial Peptides in the Pathogenesis of Atopic Dermatitis
ABSTRACT Atopic dermatitis (AD) is a chronic inflammatory skin disorder characterized by barrier dysfunction, immune dysregulation, and microbial dysbiosis. Recent studies have highlighted the multifaceted roles of antimicrobial peptides (AMPs) both as innate defenders against microbial invasion and as regulators of immune responses and skin barrier ...

Pathogenesis of Vitiligo: Integrating Immune and Non‐Immune Cell Crosstalk
ABSTRACT Vitiligo is an acquired autoimmune disease characterized by depigmented macules resulting from melanocyte loss. It is a complex multifactorial disorder in which genetic predisposition is combined with environmental factors; however, its detailed etiology remains unclear.

Distinct Roles of IL‐4, IL‐13, and IL‐22 in Human Skin Barrier Dysfunction and Atopic Dermatitis
This study reports the distinct effects of IL‐4, IL‐13, and IL‐22 on bio‐stabilized human skin with intact barriers and immune cells. IL‐4, IL‐13, and IL‐22 upregulate inflammatory mediators, disrupt skin barrier integrity, and reciprocally modulate IL‐4Rα and IL‐22Rα1 receptors.

Single Cell Analysis Reveals Dynamic Changes of Distinct Cell Populations in Human Nickel Allergy
We investigated cell types mediating the early and late inflammatory responses in human Ni2+‐allergy by scRNA‐Seq. Early responsiveness was mainly mediated by TLR4+TAGLN− endothelial cells indirectly activating TLR4−KRT16+ keratinocytes, and CCR7+DCN+ DCs. Late responsiveness was crucially driven by infiltration of KLF2+ central memory T cells and TLR4+

Toll-Like Receptor 2-Deficient Mice Are Highly Susceptible to Streptococcus pneumoniae Meningitis because of Reduced Bacterial Clearing and Enhanced Inflammation [PDF]
Toll-like receptor-2 (TLR2) mediates host responses to gram-positive bacterial wall components. TLR2 function was investigated in a murine Streptococcus pneumoniae meningitis model in wild-type (wt) and TLR2-deficient (TLR2−/−) mice.

Fibrinolysis protease receptors promote activation of astrocytes to express pro-inflammatory cytokines. [PDF]
BACKGROUND:Astrocytes contribute to the crosstalk that generates chronic neuro-inflammation in neurological diseases; however, compared with microglia, astrocytes respond to a more limited continuum of innate immune system stimulants.
