A Natural TMEM16A Inhibitor with Conformational Lock Mechanism: Thymol Reverses Secretory Diarrhea via Chloride Flux Regulation and Antiinflammatory.

Journal of Agricultural and Food Chemistry
Secretory diarrhea is characterized by excessive production of electrolytes and water by intestinal epithelial cells, resulting in persistent watery stools. This study suggested that abnormal activation of TMEM16A might contribute to its pathogenesis.

Recent advances in TMEM16A: Structure, function, and disease

Journal of Cellular Physiology, 2018
AbstractTMEM16A (also known as anoctamin 1, ANO1) is the molecular basis of the calcium‐activated chloride channels, with ten transmembrane segments. Recently, atomic structures of the transmembrane domains of mouse TMEM16A (mTMEM16A) were determined by single‐particle electron cryomicroscopy.

Involvement of TMEM16A/ANO1 upregulation in the oncogenesis of colorectal cancer

Biochimica et Biophysica Acta (BBA) - Molecular Basis of Disease, 2022
The Ca2+-activated Cl- channel ANO1 is widely expressed in epithelial cells, and ANO1 upregulation is implicated in the oncogenesis of many epithelium-originated cancers. However, whether ANO1 plays a causal role in the tumorigenesis of colorectal cancer remains largely unknown. Here, we show that ANO1 channel protein is upregulated in human colorectal

Repositioning lidocaine as a TMEM16A Ca2+-activated Cl- channel blocker for the treatment of pulmonary arterial hypertension.

Journal of Pharmacology and Experimental Therapeutics
TMEM16A forms a Ca2+-activated Cl- (ClCa) channel that plays essential roles in the cardiovascular, gastrointestinal, and central nervous systems. Dysregulation of TMEM16A expression has been implicated in the development of several diseases, making ...

Relationship between TMEM16A/anoctamin 1 and LRRC8A

Pflügers Archiv - European Journal of Physiology, 2016
TMEM16A/anoctamin 1/ANO1 and VRAC/LRRC8 are independent chloride channels activated either by increase in intracellular Ca(2+) or cell swelling, respectively. In previous studies, we observed overlapping properties for both types of channels. (i) TMEM16A/ANO1 and LRRC8 are inhibited by identical compounds, (ii) the volume-regulated anion channel VRAC ...

TMEM16A regulates MUC5AC production in airway diseases: A comprehensive review.

Current opinion in pharmacology (Print)
Abnormal secretion of the airway mucin MUC5AC is a key pathological feature in the pathologic process of airway diseases, overproduction of MUC5AC leads to airway mucus accumulation and airway obstruction, which further triggers respiratory dysfunction ...

Regulation of ANO1/TMEM16A anion permeability

Pancreatology, 2013
Background/aim: Bicarbonate (HCO3 ) is an important ingredient in many biological fluids including saliva. However, calcium activated chlo ride channels (CaCCs) in salivary gland acinar cells are known to be poorly permeable to HCO3 in most cases. Methods: To elucidate the possibility of ANO1 as a HCO3 channel, anion currents were measured in HEK 293T ...

Ursolic acid, an inhibitor of TMEM16A, co-loaded with cisplatin in hydrogel drug delivery system for multi-targeted therapy of lung cancer.

International Journal of Biological Macromolecules
The efficacy of single chemotherapy drugs in cancer treatment is often limited. Combining administration targeting multiple targets has emerged as an effective strategy to improve cancer treatment. Ursolic acid, a triterpenoid compound in various natural
