Using Synthetic Glycans to Investigate Anti-Glycan Antibodies and Explore Their Medical Potential. [PDF]
Anti‐glycan antibodies are essential in health and disease. Access to novel glycan structures paves the way for progress in antibody profiling for biomarker discovery, antibody development, and vaccine design. We summarize the strategies to synthesize and utilize synthetic glycans for the development and application of anti‐glycan antibodies in basic ...

Single CAR-T cell treatment controls disseminated ovarian cancer in a syngeneic mouse model
Background Treatment of some blood cancers with T cells that express a chimeric antigen receptor (CAR) against CD19 have shown remarkable results. In contrast, CAR-T cell efficacy against solid tumors has been difficult to achieve. Methods To examine the ...

Retinal S-antigen Th1 cell epitope mapping in patients with Behcet's disease [PDF]
Background - Retinal S-antigen (S-Ag) is a most characterized autoantigen of autoimmune uveitis. The recognized immunodominant epitope of human S-Ag in patients with uveitis has not been identified. In this study, we selected certain patients with active

Efficient chemoenzymatic synthesis of sialyl Tn-antigens and derivatives [PDF]
An N-terminal and C-terminal truncated recombinant α2-6-sialyltransferase cloned from Photobacterium sp. JH-ISH-224, Psp2,6ST(15-501)-His(6), was shown to be an efficient catalyst for one-pot three-enzyme synthesis of sialyl Tn (STn) antigens and derivatives containing natural and non-natural sialic acid forms.

Mucin-type O-glycosylation is initiated by the UDP-GalNAc polypeptide:N-acetylgalactosaminyltransferase (GalNAc-T) family of enzymes. Their activity results in the GalNAc α1-O-Thr/Ser structure, termed the Tn antigen, which is further decorated with ...

The crystal structure of a plant lectin in complex with the Tn antigen [PDF]
The structure of the tetrameric Vicia villosa isolectin B4 (VVLB4) in complex with a cancer antigen, the Tn glycopeptide (GalNAc‐O‐Ser), was determined at 2.7 Å resolution. The N‐acetylgalactoside moiety of the ligand binds to the primary combining site of VVLB4 in a similar way as observed for other Gal/GalNAc‐specific plant lectins.
