Transgenes and their contributions to epigenetic research

open access: yes, 2013
Shortly after gene transfer technologies had been established for different plant species, the first reports emerged about transgenes showing unexpected segregation patterns due to unstable expression.

Insertion of PATC-rich C. elegans introns into synthetic transgenes by golden-gate-based cloning

open access: yes, 2021
Transgenes are particularly prone to epigenetic silencing in the C. elegans germline. Here, we describe a protocol to insert introns containing a class of non-coding DNA named Periodic An/Tn Clusters (PATCs) into synthetic transgenes.

Microenvironmental Reprogramming by 3D Anisotropic Cardiac Extracellular Matrix Induces Nuclear Remodeling and Epigenetic Maturation of Chemically Induced Cardiomyocytes

open access: yesAdvanced Functional Materials, EarlyView.
A 3D anisotropic hydrogel derived from heart extracellular matrix guides cytoskeletal alignment and nuclear remodeling in reprogrammed cardiomyocyte‐like cells. This study reveals how matrix alignment modulates nuclear envelope dynamics and chromatin state, triggering transcriptional and functional maturation.

Zasp52 transgenes localize to IFM Z-discs.

open access: yes, 2016
(A) Cartoon of Zasp52 transgenes. PK and PR refer to the Zasp52 isoform the transgenes are based on (see FlyBase). Flag, Flag-tag; FSH, Flag/Streptavidin/His-tag; ZM, Zasp-like motif; a.a., amino acids.

Adipocyte‐Derived Leptolin Enhances Energy Expenditure and Prevents Obesity

open access: yesAdvanced Science, EarlyView.
We identified a novel adipokine, which we named leptolin. In humans, leptolin levels in white adipose tissue were positively correlated with exercise and negatively associated with body mass index. We observed elevated leptolin in serum from athletes and lower leptolin in serum from obese individuals.

A Rare Allele of ST5 From Wild Rice Enhances Salt Tolerance in Rice

open access: yesAdvanced Science, EarlyView.
A novel salt‐tolerance gene ST5W, characterized by a unique 36‐bp promoter insertion, is found exclusively in a small subset of Oryza rufipogon and is absent in cultivated rice. Field trials confirm ST5W significantly enhances rice yield across diverse genetic backgrounds under saline conditions.

Single-copy insertion of transgenes in Caenorhabditis elegans

open access: yes, 2008
Udgivelsesdato: 2008-NovAt present, transgenes in Caenorhabditis elegans are generated by injecting DNA into the germline. The DNA assembles into a semistable extrachromosomal array composed of many copies of injected DNA.

A Toolkit for Targeted Neuromodulation of Striatal Direct Pathway Neurons Rescues Parkinsonian Motor Deficits in Mice

open access: yesAdvanced Science, EarlyView.
An adeno‐associated virus (AAV) toolkit enables selective anatomical and functional targeting of striatal D1‐MSNs through retrograde transduction. Enhanced capsids and engineered enhancers drive robust transgene expression across murine and primate models.

mpeg1 promoter transgenes direct macrophage-lineage expression in zebrafish

open access: yes, 2011
Macrophages and neutrophils play important roles during the innate immune response, phagocytosing invading microbes and delivering antimicrobial compounds to the site of injury.
