Fluorescence detection of the transglycosylation activity of amylosucrase

Analytical Biochemistry, 2017
The purpose of this study was to investigate the novel fluorescence-based assay for the transglycosylation activity of amylosucrase (ASase). The transglycosylation activity of ASase from Deinococcus geothermalis (DGAS), ASase from Neisseria polysaccharea (NPAS), and DGAS-B (chimeric ASase wherein the B domain from DGAS was exchanged with the B domain ...

Transglycosylation by barley α-amylase 1

Journal of Molecular Catalysis B: Enzymatic, 2011
Abstract The transglycosylation activity of barley α-amylase 1 (AMY1) and active site AMY1 subsite mutant enzymes was investigated. We report here the transferase ability of the V47A, V47F, V47D and S48Y single mutants and V47K/S48G and V47G/S48D double mutant AMY1 enzymes in which the replaced amino acids play important role in substrate binding at ...

Transglycosylation of β-D-RIbofuranosylindazoles

Nucleosides and Nucleotides, 1995
Abstract Irreversible, acid-catalyzed 2 → 1 transglycosylation reactions of fully acetylated β-D-ribofuranosylindazoles were studied applying HPLC analysis. Results so obtained support the intermolecular mechanism of transglycosylation via a 1, 2-diglycosylindazole intermediate and are compared to those of the purine series.

Inhibition of Transglycosylation Involved in Bacterial Peptidoglycan Synthesis

Current Medicinal Chemistry, 2000
The continuing specter of resistance to antimicrobial agents has driven a sustained search for new agents that possess activity on drug resistant bacteria. Although several paths are available to reach this goal, the most generalized would be the discovery and clinical development of an agent that acts on a new target which has not yet experienced ...

Transglycosylation Activity ofDictyoglomus thermophilumAmylase A

Bioscience, Biotechnology, and Biochemistry, 2004
Amylase A from Dictyoglomus thermophilum is a thermophilic enzyme and has about 40% identity with 4-alpha-glucanotransferase (GTase) from Thermococcus litoralis, and both of these enzymes belong to family 57 glycosyl hydrolase. Since the transglycosylation activity of T.

Transglycosylation reactions of Thermotoga maritima α-amylase

Enzyme and Microbial Technology, 2010
Abstract The capacity of Amy A, a α-amylase from the hyperthermophilic bacteria Thermotoga maritima to carry out transfer reactions in addition to hydrolysis was investigated. Amy A is a saccharifying enzyme that catalyzes the hydrolysis of internal α-1,4 linkages of glucose polymers generating low molecular weight products (G1–G7); however, in the
